BD™ Phosflow Technologyフローサイトメーターによる細胞内リン酸化タンパク質の解析

細胞内シグナル伝達に関わっているタンパク質の活性化にはそのタンパク質に含まれているセリン/スレオニン、チロシン残基のリン酸化が大きく関わっており、様々なキナーゼやホスファターゼによって制御されています。

リン酸化タンパク質は、T細胞/B細胞シグナリング、アポトーシス、細胞成長コントロール、細胞周期、細胞骨格の再構成、転写などの重要な細胞プロセスで中心的な役割を果たしています。細胞内リン酸化タンパク質の検出は従来、ウェスタンブロッティング、免疫沈降、免疫組織染色などの技法が汎用されていますが、これらの技法の場合、解析時間がかかる、1つのサンプルで多項目の解析が難しい、比較的多くのサンプルを必要とするなどの限界がありました。

2002年、米国スタンフォード大学のギャリー・ノーラン博士の研究室より、“フローサイトメーターを用いた細胞内リン酸化タンパク質の解析”が発表されました。BDではマルチカラー解析も可能なこの新しい技法を“BDTM Phosflow Technology”としてご紹介致します。

• ウェスタンブロッティング法よりも操作が簡便、測定時間短縮

• 細胞表面抗原用の蛍光標識抗体と組み合わせるなどのマルチカラー解析を実現

図1 状態の異なる細胞集団におけるリン酸化の解析サンプル1：非リン酸化状態、サンプル2およびサンプル3：リン酸化状態サンプル2と3について、ウェスタンブロッティングでは大きな違いは認められませんが、フローサイトメーターではCでみられるようにリン酸化状態の違いを異なるパターンとして結果を得ることができます。

解析例1 ヒト末梢血単核細胞をIL-6およびIL-4で刺激した場合の解析図抗ヒト CD3 PerCP-Cy5.5、抗ヒト CD4 PE、抗ヒトリン酸化Stat6(pY641) Alexa Fluor® 488、抗ヒトリン酸化Stat3(pY705) Alexa Fluor® 647で染色後フローサイトメーターで解析。これらの解析図より、CD3+/CD4+、CD3+/CD4–、CD3–/CD4–の細胞集団においてStat6がリン酸化されていることがわかります。

解析例2 C57BL/6 マウス脾臓細胞をPMA+LPS で刺激した場合の解析図抗マウス CD11b PerCP-Cy5.5、抗マウス CD45R/B220 Alexa Fluor® 488、抗マウスリン酸化p38 PE、抗マウスリン酸化ERK1/2 Alexa Fluor® 647 で染色後フローサイトメーターで解析。これらの解析図より、p38は主にCD11b+/B220– 細胞集団が刺激により活性化されリン酸化されていることがわかりますが、一方ERK1/2ではどの細胞集団においてもリン酸化されていることがわかります。

解析例2を得るために、実施した Protocol （概略）

サンプル、リン酸化タンパク質、同時に測定したい細胞表面抗原などによって周辺試薬（細胞固定試薬、細胞膜透過試薬）の組み合わせが異なる場合があります。（詳しくはアプリケーションハンドブックをご覧ください。）

マウス脾臓細胞を PMA+LPSを用いて15分間刺激

BD PhosflowTM Lyse/Fix Buffer (Cat. No. 558049)を用いて細胞を固定（37℃10分間）

　　　　　　　　　　　　　　　　　　

BD PhosflowTM Perm Buffer III (Cat. No. 558050)を用いて細胞膜透過処理（氷上30分間）

BD Fc BlockTM (Cat. No. 553141)を用いて、Fc Receptor をブロック（氷上15分間）
