ABSTRAK

Gel oftalmik adalah sediaan oftalmik yang dimaksudkan untuk memberikan waktu kontak yang lebih lama dengan mata sehingga dapat meningkatkan bioavailabilitas obat. Telah dilakukan pengembangan sediaan gel oftalmik yang bertujuan mengetahui formula dengan zat aktif kloramfenikol 0,5% dengan basis gel kombinasi hidroksi propil metil selulosa dan karbomer sebesar (0:1, 1:0, 2:1, 4:1). Pembuatan formula dilakukan melalui tahap pembuatan basis, pencampuran dengan zat aktif, dan sterilisasi menggunakan autoklaf pada suhu 121C selama 15 menit. Hasil evaluasi yang meliputi organoleptik, pH. viskositas, dan kadar zat aktif menunjukkan bahwa F4 adalah formula yang terbaik. F4 memiliki konsistensi encer, jernih dan tidak berbau, pH sebesar 7,49, viskositas sebesar 55 cP, dan kadar zat aktif sebesar 100%. Pada uji pengamatan sediaan gel oftalmik selama 28 hari penyimpanan menunjukkan tidak terjadi perubahan organoleptik, terjadi perubahan pH dan viskositas namun masih dalam rentang persyaratan, juga terjadi penurunan kadar zat aktif sekitar 52,82%-53,66%. Hasil uji aktivitas antibakteri sediaan menunjukkan bahwa sediaan gel oftalmik efektif terhadap bakteri uji Staphylococcus aureus.Kata kunci: gel oftalmik, kloramfenikol, carbopol, HPMCABSTRACTOphthalmic gel is an ophthalmic preparation intended to provide a longer contact time with the eye to increase the bioavailability of the drug. ophthalmic gel has been developed to discover the formula contained chloramphenicol 0.5% as active ingredients with the combination of gel hydroxy propyl methyl cellulose, and carbomer of (0:1, 1:0, 2:1, 4:1) as the base. formula was made by several stages which were mixing with the active ingredient, and sterilization using autoclave at 121 C for 15 minutes. Results which covered organoleptic evaluation, pH. viscosity, and the levels of the active substance showed that F4 was the best formula. F4 had a watery consistency, clear and odorless, pH of 7.49, viscosity of 55 cP, and the levels of the active substance at 100%. test observations ophthalmic gel for 28 days of storage showed no changes in the organoleptic. PH and viscosity changes occured, but still within the range of requirements, also levels of the active substance was decreased around 52.82% -53.66%.
