1. FINES Y OBJETIVOS
El Instituto Universitario de Bio-‐Orgánica Antonio González (IUBO-‐AG) es un centro multidisciplinar
orientado a la investigación en el campo de la Química Bio-‐Orgánica, Productos Naturales Bioactivos y Química Médica. El Instituto fue fundado en los años 60 con la necesidad de estudiar los metabolitos secundarios producidos por organismos marinos y terrestres, para proporcionar una nueva fuente de compuestos únicos. Actualmente, los objetivos del Instituto son el aislamiento, la biosíntesis, los cultivos de microorganismos, la biotecnología y la síntesis total de sustancias farmacológicamente activas procedentes de fuentes naturales. Además, son áreas de investigación de interés actual la evaluación biológica, el aislamiento y producción de toxinas, estudios NMR de procesos biológicos, insecticidas y repelentes naturales, etc.
Líneas de investigación IUBO-‐AG • Agentes anticancerígenos procedentes de fuentes naturales Varios productos naturales y sus derivados semisintéticos son producidos dentro de varios
proyectos comunes para el desarrollo de antitumorales con nuevos mecanismos de acción. Algunos productos recientemente han mostrado in vitro una reversión de la MDR (multirresistencia a fármacos) y efectos quimiopreventivos sobre el virus de Epstein-‐Barr.
• Estudios de la relación entre Estructura-‐Actividad Para entender la base estructural de la actividad biológica de una molécula y para dirigir el diseño
de un fármaco más potente, el modelo cuantitativo tridimensional de la relación de la estructura-‐actividad (3D-‐QSAR) se realiza a partir de quimiotecas de compuestos relacionados.
• Agentes antimicrobianos procedentes de fuentes naturales Productos naturales procedentes de plantas y de derivados semisintéticos son de interés para
luchar contra varios microorganismos y sus mecanismos de acción. • NMR en el estudio de procesos biológicos Técnicas avanzadas de Resonancia Magnética Nuclear (NMR) se aplican para determinar
interacciones enzima-‐inhibidor como una alternativa novedosa en la búsqueda de nuevos fármacos. • Organismos marinos y toxinas Los dinoflagelados son organismos marinos responsables de las mareas rojas y el envenenamiento
de moluscos. Entre otras, el ácido okadaico y las yessotoxinas son las toxinas más comunes presentes en crustáceos europeos. Desarrollamos sistemas de cultivos para proveernos de suficientes cantidades de toxinas para realizar estudios biológicos.
• Insecticidas y repelentes Se están aislando productos naturales para su uso contra plagas, especialmente las que afectan a
la agricultura. Estos proyectos se llevan a cabo en colaboración con instituciones públicas y compañías agroquímicas de Europa y de América Latina.
• Productos químicos y farmaquímicos Nuestro instituto cuenta con una larga experiencia en el campo de la síntesis orgánica destinada a
la síntesis de sustancias medicinales, especialmente enfocada en procesos asimétricos. Es de interés particular encontrar el desarrollo de nuevas metodologías para la síntesis total de sustancias activas biológicamente como toxinas, aminoácidos, análogos de la esfingosina, alcaloides, etc.

• Catálisis metálica sostenible En el actual contexto socioeconómico, el concepto de sostenibilidad ha irrumpido con mucha
fuerza. Para el ser humano, la sostenibilidad consiste en satisfacer las necesidades de la actual generación sin sacrificar la capacidad de futuras generaciones. Dicho concepto tiene implicaciones medioambientales, económicas y sociales.
Actualmente se consumen muchos recursos, se encarecen los precios, mientras la sociedad demanda procesos más respetuosos con el medio ambiente.La catálisis posee el potencial para convertirse en una de las claves tecnológicas del siglo XXI. Así se podrán llevar a cabo nuevas reacciones y se desarrollarán procesos que permitan un ahorro de energía. Se podrán desarrollar nuevas metodologías sintéticas que tengan el menor impacto medioabmiental. Sin embargo muchos de los catalizadores empleados hoy en día son metales de transición caros, poco abundantes en cantidades que garanticen un total suministro en épocas venideras. Dentro de este contexto, estamos interesados en el desarrollo de nuevas metodologías sintéticas orientadas a la obtención de heterociclos (oxigenados y nitrogenados) de diversos tamaños, con potencial actividad biológica. Aplicar dicha metodología en la síntesis de productos naturales bioactivos y derivados utilizando una catálisis metálica sostenible. Por todas estas razones el hierro es una excelente alternativa a los metales preciosos en catálisis. Es uno de los metales más abundantes, es accesible, con bajo precio y respetuoso con el medio ambiente. Varias sales de hierro y complejos de hierro son accesibles comercialmente a gran escala o fáciles de sintetizar. Es importante destacar que el hierro forma parte de sistemas biológico como elemento esencial. El incremento en el número de reacciones catalizadas por hierro que se han publicado reflejan una mayor demanda de la química sostenible.
• Descubrimiento fenotípico de compuestos antiproliferativos Nuestra aproximación se fundamenta en el uso de estrategias convergentes para la identificación
precoz de la diana terapéutica de cada molécula bioactiva mediante la aplicación de una estrategia global. Los compuestos que se analizan provienen no solo de grupos de investigación de la Universidad de La Laguna, sino de colaboraciones estables que mantenemos con grupos químicos nacionales e internacionales.
• Farmacología e investigaciónes biomédicas
El IUBO-‐AG participa activamente en programas de investigación en colaboración con los hospitales y centros de investigación biomédicos de Canarias. El resultado de esta iniciativa es la constitución del CIBITEN (Centro de Investigación Biomédica de Tenerife) diseñado hacia el desarrollo en conjunto en los distintos aspectos de las Ciencias Biomédicas y de la Salud en Tenerife y Canarias.

8. PROYECTOS DE INVESTIGACIÓN SUBVENCIONADOS
Síntesis de nuevas entidades químicas para cartografiar la bioactividad en el espacio químico Resumen Nuestra propuesta de investigación se centra en la síntesis de pequeñas moléculas para cartografiar la bioactividad en el espacio químico. El objetivo general del proyecto es diseñar y desarrollar metodologías sintéticas basadas en procesos catalíticos (orgánicos y metálicos) y/o dominó para la síntesis de colecciones moleculares de moléculas pequeñas, las cuales incluirán productos naturales y homólogos, motivos estructurales privilegiados y plataformas moleculares sintéticas. Haremos uso de estrategias sintéticas tales como síntesis orientada a una estructura (producto natural) (TOS), síntesis de estructuras homólogas a partir de intermedios avanzados comunes (DTS) y síntesis orientada a la diversidad como ejes de innovación para la construcción molecular y el descubrimiento de nuevos cabezas de serie. Las colecciones de pequeñas moléculas serán evaluadas de acuerdo a nuestra implementación de la aproximación denominada “Phenotypic Drug Discovery” (descubrimiento de fármacos basados en fenotipos), la cual permite explorar contextos de relevancia biológica sin conjeturar mecanismos de acción. Los módulos existentes de evaluación contribuirán a explorar áreas relevantes del espacio farmacológico (biológico) (antitumorales, antibióticos, anti-‐infamatorios), en busca de nuevas topologías químicas bioactivas. Utilizaremos los tres módulos de ensayos fenotípicos denominados Cáncer, Enfermedades Infecciosas y Lesión Pulmonar Aguda. Los cabezas de serie seleccionados en los ensayos primarios serán validados mediante ensayos secundarios y de confirmación para descubrir sus mecanismos de acción y así como la identificación de la diana biológica. En el contexto de este proyecto desarrollaremos un nuevo módulo de evaluación destinado al estudio de la actividad “antibiofouling” (el biofouling es la acumulación no deseada de depósitos, esencialmente microbiológicos, sobre una superficie artificial sumergida o en contacto con agua de mar). Synthesis of Novel Chemical Entities to Map Bioactivity In The Chemical Space Synocemabics Summary This research proposal relates to the synthesis of small molecules to map bioactivity in the chemical space. The general aim of the project is to design and develop synthetic methodologies based on domino and/or catalytic manifolds (organic and metallic) for the synthesis of libraries of small molecules including natural products, natural products-‐like, privileged structural motives and synthetic scaffolds. We will make use of front edge synthetic strategies such as target-‐oriented synthesis, diverted–oriented synthesis and diversity-‐oriented synthesis as innovation axes for molecular construction and lead discovery. The resulting small molecule libraries will be screened using our implementation of the so-‐called Phenotypic Drug Discovery approach, which allows interrogating relevant biological contexts without predisposed bias toward action mechanisms. modules will contribute to explore relevant areas of the pharmacological (biological) space (antitumor, antibiotics, anti-‐inflammatory) in the search for novel bioactive chemical topologies. Three phenotypic assay modules, namely Cancer, Infectious Diseases and Acute Lung Injury will be used. Primary assays will be used to select initial lead compounds that will be submitted to secondary and confirmatory assays to unravel their mechanism of action in addition to identification of the biological target. Dinoflagelados tóxicos en las costas españolas: Estudios químico, biológico y de modos de acción. Producción de metabolitos de interés y evaluación de su potencial biotecnológico Resumen El crecimiento incontrolado de especies tóxicas de dinoflagelados (mareas rojas tóxicas), genera un tremendo impacto en la salud pública a través del envenenamiento de moluscos y su paso a la cadena alimentaria. Sin embargo este no es el único riesgo al que se ve sometida la población que entra en contacto con el fenómeno. En las aguas costeras europeas, hay un grupo de toxinas importantes de naturaleza policétida, en su mayoría liposolubles y de elevado grado de toxicidad. Estas potentes sustancias bioactivas han sido detectadas en su mayoría en los últimos años y presentan estructuras no encontradas en otros organismos marinos o terrestres por lo que, en general, poseen mecanismos de acción novedosos. Todo ello genera la necesidad del estudio integral del fenómeno: biología de los microorganismos responsables, incluyendo las condiciones para su cultivo artificial; descubrimiento de nuevas estructuras y biogénesis de las mismas; establecimiento del modo de acción biológica de las nuevas sustancias y establecimiento de las condiciones para la producción masiva de los microorganismos productores de los metabolitos de interés. El grupo de Vigo estudiará las cepas de Ostreopsis responsables de fenómenos de intoxicación respiratorias en costas italianas y españolas y evaluará los riesgos asociados a este problema. Asimismo estudiará las cepas de Gambierdiscus toxicus aisladas en las Islas Canarias relacionadas con el primer caso de intoxicación ciguatérica en esta parte del Atlántico por consumo de pescado en Fuerteventura. El grupo de La Laguna, estudiará los metabolitos generados por el Prorocentrum belizeanum, que incluyen análogos del AO sin su estructura característica, policétidos de elevado peso molecular y naturaleza macrólida, poseedores de un alto potencial biológico. Asimismo, se estudiarán químicamente las nuevas cepas de Ostreopsis

productoras de ostreocinas. Los conocimientos acerca de los efectos que causan sobre el SNC estas ficotoxinas son muy limitados. Sin embargo, los datos previamente obtenidos por el grupo de Oviedo, justifican el uso de cultivos neuronales tanto para estudiar su mecanismo de acción como para las actividades biológicas de los compuestos suministrados por los otros grupos. El grupo de Almeria, añade los aspectos tecnológicos para tratar de resolver los problemas asociados con el cultivo masivo de células de dinoflagelados. Hasta donde sabemos, no existe otro grupo en el mundo en ingeniería de bioprocesos dedicado a desarrollar metodología para estos fines. Toxic Dinoflagellates in the Spanish Coast: Chemistry, Biology and Modes of Action. Production of Interesting Metabolites and Evaluation of their Biotechnological Potential Summary The uncontrolled growth of toxic dinoflagellates (toxic red tides), can have novice effects of paramount importance on public health through the shellfish consumption and its subsequent transition into the food chain. However, there are additional risks for the population that comes in contact with this phenomenon. In European coastal waters, there is an important group of highly toxic compounds which posses mainly liposoluble polyketide nature. These potent bioactive substances have been detected principally in recent years showing novel both structures and mechanism of action not found in other marine or terrestrial organisms. These discoveries generate the need for comprehensive studies of the phenomena: biology of the responsible organisms, including the conditions for artificial cultures; discovery of new structures and their biogenesis; determination of the mode of action of those novel biological substances as well as the conditions necessary for their production using large-‐scale cultures of the micro-‐organisms. The group of Vigo will study strains of Ostreopsis responsible for respiratory poisoning events in Italian and Spanish coasts and will also assess the risks associated with this problem. In addition it will study Gambierdiscus toxicus strains isolated in the Canary Islands associated with the first case of ciguateric poisoning in this part of the Atlantic caused by fish consumption in Fuerteventura. The group of La Laguna, will study the metabolites generated by Prorocentrum belizeanum, including analogues of OA showing important structural differences and high molecular weight polyketide compounds with macrocyclic structures, that show high biological potential. In addition, new toxic strains of Ostreopsis, will be studied chemically. As far as we know, very limited information is available about the effects of these toxins to the CNS. The results obtained by the group of Oviedo, fully justify the use of these cultures for studying the mechanisms of action of these compounds, as well as for the screening of biological activities of new dinoflagelate-‐produced molecules forwarded by the other groups. The commercial potential of the toxins and other bioactive substances produced by marine dinoflagellates is hindered by the enormous difficulty to undertake massive cultures of dinoflagellates. The group of Almería will add the necessary biotechnological expertise into the project to tackle those problems involved in the massive culture of dinoflagellates. To our best knowledge there is not a single research group over the world in the field of Bioprocess Engineering involved in the development of a bioprocess for obtaining metabolites of interest from marine dinoflagellates. Compuestos Bioactivos de Organismos Marinos: estudios estructurales, farmacológicos y biotecnológicos Resumen Las microalgas marinas juegan un papel crucial en los ecosistemas debido a su actividad fotosintética y de entre ellas, nuestro proyecto se interesa por el grupo de los dinoflagelados capaces de originar afloramientos o “blooms” conocidos como “mareas rojas”. Éstas se clasifican habitualmente en función de los efectos toxicológicos en humanos. Por otro lado, los dinoflagelados marinos podrían llegar a ser auténticas “factorías celulares” de importantes sustancias bioactivas con diferentes aplicaciones farmacológicas. Estudiaremos a gran escala especies de Prorocentrum orientados a los metabolitos más polares, apoyándonos en los hallazgos en el IUBO de Belizeanolido y Ácido Belizeanóico, dos ejemplos de los denominados “SuperCarbonChain compounds (SCC)” y otros ejemplos de lactonas macrocíclicas. Dado el interés de estos dinoflagelados como productores, en la UAL se propone establecer las condiciones para su cultivo semi-‐industrial y productivo. El objetivo es adaptar y poner a punto fotobiorreactores de decenas de litros trabajando el escalado para la máxima producción de toxinas y de biomasa en el cultivo externo para los dinoflagelados seleccionados. Moléculas de esta complejidad generan nuevos problemas que han de resolverse. En este sentido se está desarrollando una metodología por RMN directa y simple para establecer la configuración de los centros quirales en sistemas de anillos de cinco miembros muy comunes en este tipo de estructuras y de establecer el origen biosintético de estas estructuras. El interés de estas sustancias también radica en su potente actividad inhibitoria de proteínas fosfatasas, y hoy en día sigue suponiendo un gran reto la búsqueda de nuevos inhibidores selectivos. En el IUBO se realizarán estudios de inhibición in vitro; estudios estructurales y conformacionales del AO y análogos; cálculos teóricos de docking para determinar su modo de unión a los enzimas y establecer los requisitos estructurales necesarios para la inhibición selectiva de PP1 y PP2A. Los datos de inhibición in vitro serán contrastados en la UO con los de cultivos de cerebelo, para identificar los motivos estructurales asociados a la efectividad inhibidora en células vivas, la cual, dada la gran complejidad estructural de las PPs y su regulación, puede diferir significativamente de la actividad in vitro. Se estudian también otros poliéteres de algas rojas del género Laurencia para información de moléculas y motivos estructurales más simples con actividad inhibitoria. En la UAL, se estudian sus interacciones con células inmunitarias, evaluando la toxicidad y la función biológica de estas células ante la toxina o bioproducto. En el caso de linfocitos T, su capacidad para reconocer inmunógenos. Dada la gran sensibilidad del SNC a la acción a largo plazo de estos compuestos usaremos cultivos celulares primarios y organotípicos de SNC. Las propiedades eléctricas neuronales permiten aproximaciones únicas de determinación de efectos fisiológicos subtóxicos como los registros extracelulares con matrices de microeléctrodos. El estudio de las vías bioquímicas estará orientado especialmente a la identificación de mecanismos neuroprotectores. Finalmente en este proyecto nos planteamos iniciar el estudio a pequeña escala de 50 especies de diferentes géneros de dinoflagelados de la colección del IEO (Vigo) para estudiar su bioactividad y elegir los adecuados para proseguir su escalado y futuros estudios

Bioactive Molecules From Marine Organisms: Structural, Pharmacological And Biotechnological Studies Summary Among marine organisms, microalgae play a crucial role in ecosystems due to their photosynthetic activity. This group is characterized by their ability to produce blooms known as "red tides” and classified according to the toxicological effects provoked in humans. There is no doubt that marine dinoflagellates could become real "cell factories" of important bioactive substances with different biotechnological applications. Thus, on the basis of the results recently obtained from of P. belizeanum, in this project we propose to continue the development of large-‐scale cultures of Prorocentrum species focussed on the search of highly polar metabolites. Certainly, we have isolated and characterized two new examples of the so-‐called "SuperCarbonChain compounds,SCC)" named belizeanoic acid and belizeanolide. Furthermore we have detected similar metabolites that are still under study. In addition, we propose to undertake in the UAL further studies to find the optimal the conditions to produce massive cultures. The ultimate goal of our project would be to adapt and develop the culture of dinoflagellates in external photobioreactors. It is obvious that complex molecules create new problems to be solved. On our side, we are developing a simple NMR methodology to establish the configuration of stereogenic centers in five-‐membered rings, which are common in this kind of compounds. This approach includes the synthesis of models that will be used to confirm theoretical approaches and we also try to establish their biosynthetic origins. In addition, the interest of this group of substances is mainly based on their potent biological activities that include new modes of action. Regarding the inhibitory activity of protein phosphatases, the design of this kind of selective inhibitors is the main challenge in this field nowadays. Therefore, we have undertaken structural and conformational studies in solution of AO and analogues and theoretical docking calculations to determine a plausible binding mode. These studies will continue aiming to find simplified molecules fulfilling the necessary structural requirements to achieve a selective inhibition of PP1 and PP2A. Data from in vitro inhibitory activity obtained at IUBO will be compared and complemented with the evaluation of biochemical and toxicological effects in neuronal and glial cells cultures developed in UO, which will help us to establish structure-‐activity correlations and to identify those structural motifs that determine more effective inhibitors in living cells. However, given the structural complexity of protein phosphatases and the cellular regulatory mechanisms they control it may differ significantly from the inhibitory activity determined in the test tube. Furthermore, these studies will be extended to other types of polyethers found in red algae of the genus Laurencia. The group of UAL will study the interaction between animal immune cells and the isolated compounds, evaluating both their toxicity and biological function, for instance studying the ability to recognize immunogens of T cells. UO group will analyse the effects on the CNS using in vitro systems of primary cerebellar neurons, glial cells and organotypic cultures. We will study the biochemical pathways involved in the physiological and toxicological effects and on the development of novel neuroprotective therapies. Finally in this project we plan to start small-‐scale cultures of 50 species of dinoflagellates from IEO collection to assess their bioactivity and select the new organisms for future studies. Desarrollo de nuevos procesos catalíticos dirigidos a la síntesis de moléculas bioactivas Resumen El objetivo global de este proyecto es la síntesis de moléculas pequeñas con uso potencial como sondas biológicas o fármacos. Esencialmente, diseñaremos sistemas catalíticos (orgánicos y metálicos) que se aplicarán a la síntesis de moléculas orgánicas de baja masa molecular y estructuralmente complejas. Estas moléculas incluirán tanto productos naturales como derivados activos con estructuras simplificadas (síntesis orientada a un objetivo) como cabezas de serie biológicos de baja masa molecular (síntesis orientada a la diversidad). Estos cabezas de series serán seleccionados por diseño racional basado en cálculos fiables o por evaluación de la actividad biológica en “tiempo real”. Significa que para cada compuesto sintetizado su actividad biológica será analizada en el plazo de una semana, permitiendo un desarrollo rápido y directo de los nuevos análogos para los estudios de Relación Estructura-‐Actividad (SAR). Los principales tópicos a investigar en este proyecto incluyen órgano-‐catálisis en agua, metalo-‐catálisis, síntesis modular y orientada a la diversidad de moléculas pequeñas, síntesis de productos naturales y homólogos, diseño y síntesis racional de nuevos farmacóforos e investigación de la actividad biológica en tiempo real. Development of New Catalytic Process Oriented to the Syntheis of Bioactive Molecules Summary The overall objective of this project resides on the synthesis of small molecules with potential use as biological probes or drugs. Essentially, we will design catalytic systems (organic and metallic) to be applied to the synthesis of small and complex organic molecules (skeletally-‐complex small molecules). These will range from natural products and related simplified structures (target-‐oriented synthesis) to small-‐molecule biological leads (diversity-‐oriented synthesis). These leads will be selected by rational design based on reliable computational approaches and/or by biological screening in “real time”. It means that every synthesized compound will be analyzed for its biological activity within one week, allowing a fast and direct development of new analogs for Structure-‐Activity Relationship (SAR) studies and lead optimization. The main research topics pursued in this project include organo-‐catalysis in water, metallo-‐catalysis, modular and diversity-‐oriented synthesis of small molecules, synthesis of natural products and homologues, rational design and synthesis of biological ligands and biological screening in real time. MAREX: Exploring Marine Resources for Bioactive Compounds: From Discovery to Sustainable Production and Industrial Applications 2010-‐2014 Project description Biodiversity in the seas is only partly explored, although marine organisms are excellent sources for many industrial products. Through close co-‐operation between industrial and academic partners, the MAREX project will collect, isolate and classify marine organisms, such as micro-‐ and macroalgae, cyanobacteria, sea anemones, tunicates and fish from the Atlantic, Pacific and Indian Oceans as well as from the Mediterranean, Baltic and Arabian Seas. Extracts and purified compounds of these organisms will be studied for several therapeutically and industrially significant biological activities, including anticancer, anti-‐inflammatory, antiviral and anticoagulant activities by applying a wide variety of screening tools, as well as for ion channel/receptor modulation and plant growth regulation. Chromatographic isolation of bioactive compounds will be followed by structural determination. Sustainable cultivation methods for promising organisms, and biotechnological processes for selected compounds will be developed, as well as biosensors for monitoring the target compounds. The work will entail sustainable organic synthesis of selected active compounds and new derivatives, and development of selected hits to lead compounds. The project will expand marine compound libraries. MAREX innovations will be targeted for industrial product development in order to improve the growth and productivity of European marine biotechnology. MAREX aims at a better understanding of environmentally conscious sourcing of marine biotechnology products and increased public awareness of marine biodiversity and potential. Finally, MAREX is expected to offer novel marine-‐based lead compounds for European industries and strengthen their product portfolios related to pharmaceutical, nutraceutical, cosmetic, agrochemical, food processing, material and biosensor applications. Uso de estructuras privilegiadas y dianas moleculares en eldiseño racional y eficiente de nuevas entidades como fármacos antitumorales y antiparasitarios Resumen El objetivo que perseguimos en el presente proyecto es el descubrimiento y desarrollo de nuevas moléculas con potencial actividad antitumoral o antiparasitaria. Llevaremos a cabo una aproximación altamente eficiente basada en el uso de estructuras privilegiadas, estrategias sintéticas eficaces como las reacciones dominó, estudios de modelización 3D-‐QSAR y tipo Docking, y herramientas biotecnológicas para la producción de metabolitos secundarios de interés. Las distintas quimiotecas serán focalizadas para ser ensayadas frente a dianas específicas como Jak/Stat/Socs, receptores de estrógeno, colinoquinasa, topoisomerasa II, etc. Los compuestos se ensayarán frente a las distintas dianas en función de sus esqueletos y de antecedentes previos de actividad de compuestos similares y de estudios de modelización tipo Docking cuando existan datos cristalográficos de las correspondientes dianas moleculares lo que facilitará el diseño racional de las nuevas entidades. Mejoraremos las propiedades biológicas y/o farmacológicas de moléculas bioactivas provenientes del cribado frente a distintas dianas moleculares terapéuticas, por medio de estudios de relación estructura-‐actividad (SAR) y 3D-‐QSAR. Por último nos proponemos el desarrollo de hidrogeles como sistemas portadores-‐suministradores de fármacos. Summary The main objective of the present research proposal is the discovery and development of novel molecular entities displaying antitumour or antiparasitic activities. We will carry out an efficient approach based on the use of priviliged structures, domino reactions, 3D-‐QSAR, Docking studies and biotechnological techniques for the production of bioactive secondary metabolites. The chemical libraries will be assayed against specific molecular targets such as Jak/Stat/Socs, estrogen receptors, choline kinase, topoisomerase II, etc. The compounds will be tested in the corresponding assays depending of their skeletons, bibliographic antecedents of similar structures and Docking studies which will conduct the rational design of new molecular entities. We will also try to increase the biological and pharmacological properties of the bioactive molecular through SAR and 3D-‐QSAR studies. Evaluación del contenido en estrógenos naturales y sintéticos en leche y derivados lácteos mediante técnicas analíticas avanzadas y nuevos procedimientos de extracción Resumen El análisis de estrógenos tanto naturales como sintéticos constituye un tema de especial relevancia en las muestras alimenticias, pues está ampliamente demostrado que un exceso de los mismos en las dietas producen multitud de desórdenes en los sistemas endocrinos que, en muchas ocasiones, además de producir diferentes enfermedades (obesidad, hiperactividad, diabetes, disminución de la fertilidad, etc), pueden degenerar en cáncer, sobretodo de próstata, ovario y pecho, por lo que estudiar el tipo y contenido de este tipo de compuestos en alimentos básicos constituyentes de nuestra dieta es crucial. El exceso de estrógenos y las modificaciones en el sistema endocrino están causados por una amplia variedad de factores, entre los que se pueden incluir: ingerir una dieta rica en carnes que contiene estrógenos sintéticos, tomar leche y derivados lácteos y comer alimentos envasados en cajas o embalajes plásticos. Hoy en día, el consumo de leche por niños y adultos constituye una parte muy importante en la alimentación de la humanidad, dado que supone cerca de 500 millones de toneladas/año, gracias al desarrollo y a los avances de la ganadería y, entre un 60 y 70% de la ingesta de este tipo de compuestos. Aunque existen estudios en los que se determinan los estrógenos en este tipo de alimento, éstos sólo se han hecho de cara a identificar los principales estrógenos naturales y en algunos casos los sintéticos, pero existe una laguna muy amplia en lo que respecta a la determinación de los metabolitos de los mismos (derivados metilados, hidroxilados y glicosilados), los cuales constituyen más del 85% de la ingesta. Esto ha sido debido a que los procedimientos de extracción de este tipo de productos suele ser muy complejos y tediosos, y a que a pesar de que se han producido grandes avances en el campo analítico no se han sido utilizados en el campo de análisis de alimentos, por lo que establecer métodos de extracción altamente selectivos, eficaces y rápidos utilizando nanotubos de carbono, líquidos iónicos convencionales y sintetizados en el laboratorio, microextracción en fase sólida o hollow fiber acoplados a técnicas avanzadas de análisis (fundamentalmente HPLC y CE) para su determinación, constituye un aspecto fundamental y que apenas ha sido explorado por la comunidad científica. El determinar este tipo de metabolitos así como establecer la cantidad de fuentes estrogénicas (procedentes de las leches y derivados) a las cuales está sometida la población ayudará no sólo a esclarecer los efectos que este tipo de compuestos ejerce sobre distintos tipos de enfermedades, sino que permitirá al sector lácteo realizar una evaluación eficaz de cara a la racionalización de este tipo de productos para la población, incrementando la seguridad en el consumo de dichos alimentos. Por tanto esta propuesta tiene como objetivo fundamental la determinación del contenido estrogénico (incluyendo estrógenos naturales, sintéticos, así como sus metabolitos, además de precursores de actividad estrogénica procedente de envases plásticos) en leches y derivados (yogur y quesos) mediante procedimientos de extracción altamente selectivos, económicos y rápidos para su posterior identificación y cuantificación mediante técnicas cromatográficas y electroforéticas. Se pretende aplicar dichas metodologías a un amplio abanico de muestras de leche de diferente naturaleza y con diferente contenido graso, así como a derivados lácteos (yogures y quesos en este caso fabricados en la Comunidad Autónoma Canaria) consumidas ampliamente por la población así como evaluar su

contenido estrogénico. Estos estudios (desarrollo de metodologías de extracción, determinación y evaluación) que se enmarcan dentro del campo de seguridad alimentaria, permitirán hacer evaluaciones eficaces de los contenidos de los mismos y establecer medidas preventivas de cara a disminuir los efectos de este tipo de compuestos sobre la población y así prevenir desordenes y/o enfermedades. Summary The analysis of natural and synthetic estrogens in foods constitutes a topic of special concern, since it is widely demonstrated that their excessive dietary intake is responsible for a good number of disorders of the endocrine system. Such disorders can cause different illnesses i.e. obesity, hyperactivity, diabetes, infertility, etc. but may also be responsible for the appearance of certain types of cancer (ovarian, mammalian and prostate cancer). Such estrogenic excess in the diet, as well as the modifications of the endocrine system, are caused by several factors like high ingest of meat products with synthetic estrogens, drinking milk and its derivatives, eating plastic packed foods, etc. That is why the study of the estrogenic content of basic food products is of high interest. Nowadays, children and adults consumption of milk constitute a very important part of human’s nutrition since nearly 500 millions of tons per year are worldwide consumed (thanks to modern farming practices). Such important consumption also represents the intake of 60-‐70% of these compounds in the diet. Although there already exist several studies in which the determination of estrogens in milk has been carried out, they have been mainly focused on the identification of the most important natural estrogens (and in few cases synthetic). However, few works have focused on their metabolites analysis (methylated, hydroxilated and glicosilated), which constitute 85% of the dietary intake. The main reason of this hardly explored field has probably been the fact that the extraction procedures developed up to now for their analysis are very complex and tedious. In fact, modern analytical techniques employing highly selective, efficient and quick extraction methods using new materials like carbon nanotubes, ionic liquids or hollow fibers, as well as solid-‐phase microextraction, coupled to advanced separation techniques (mainly HPLC and capillary electrophoresis), have hardly been applied in this field. Therefore, estrogenic metabolite analysis in milk products will allow establishing relationships between their high dietary intake and illnesses, but also will awake the interest of milk industries in order to rationalize the consumption of these products. The main objective of this proposal is to determine the estrogenic content (natural, synthetic and their metabolites, as well as some food packings constituents which are precursors of estrogenic activity) in milk and milk products (yoghourt and cheese) using highly selective, economic and quick extraction procedures for the ulterior analysis using cromatographic and electrophoretic techniques. Samples of different nature and fat content will be analyzed, all of them manufactured in the Canary Islands. The objective of this project is therefore focused on the food safety field. Mecanismo de Acción de Nuevos Compuestos Antitumorales Disruptores de la División Celular Resumen Esta propuesta de investigación se refiere a la farmacología molecular de nuevos compuestos antitumorales resultantes de nuestro programa de cribado basado en ensayos fenotípicos (PDD). El objetivo general de este proyecto es la evaluación a nivel molecular de pequeñas moléculas que incluyen productos naturales y sus análogos, motivos estructurales privilegiados y estructuras moleculares con un interés relevante en aquellos productos que afectan a la división celular. Se considerarán dos temas generales. A) Compuestos líderes existentes: Previamente hemos identificado diversos líderes (p.ej. el inhibidor de topoisomerasa II DTA0100) que requieren estudios mecanísticos adicionales. B) Compuestos líderes nuevos: Las librerías de pequeñas moléculas (provenientes de nuestras colaboraciones externas) serán evaluadas utilizando nuestra implementación de la aproximación PDD, que nos permite interrogar contextos biológicos relevantes sin predisposición hacia algún mecanismo de acción en particular. Tres módulos de ensayos fenotípicos desarrollados por el grupo solicitante contribuyen a explorar las áreas relevantes del espacio farmacológico (antitumoral, antibiótico, antiinflamatorio), denominados Cáncer, Enfermedades Infecciosas y Lesión Pulmonar Aguda. Los ensayos primarios se utilizarán para una selección inicial de compuestos líderes, que a su vez serán sometidos a ensayos secundarios y confirmatorios para descifrar su mecanismo de acción además de la identificación de la diana biológica. El módulo de Síntesis de Pequeñas Moléculas complementará el trabajo con todos los aspectos relacionados con el desarrollo como fármaco (escalado, propiedades de buen fármaco, etc.) que no son contemplados por los grupos de colaboradores externos. The general aim of the project is the evaluation at a molecular level of small molecules (SMs) including natural products, natural products-‐like, privileged structural motives and synthetic scaffolds with a prominent interest in those that can affect cell division. Two main topics are considered. A) Ongoing lead compounds: Previously, we identified several leads (e.g. the topoisomerase II inhibitor DTA0100) that require further mechanistic studies. B) New lead compounds: The resulting SM libraries (available from long and short-‐term external collaborations) will be screened using our implementation of the PDD approach, which allows interrogating relevant biological contexts without predisposed bias toward mechanism of action. Three phenotypic assay modules developed by the applicant group contribute to explore relevant areas of the pharmacological space (antitumor, antibiotics, anti-‐inflammatory), namely Cancer, Infectious Diseases and Acute Lung Injury. Primary assays will be used to select initial lead compounds that will be submitted to secondary and confirmatory assays to unravel their mechanism of action in addition to identification of the biological target. The Small Molecule Synthesis module will fulfill with those drug development issues (scaling up, drug-‐like properties, etc.) that are not covered by the external collaborating synthetic groups.

Desarrollo de nuevos fármacos antibacterianos y antifúngicos derivados de estructuras privilegiadas de origen marino Resumen El proyecto que se propone desarrollar es multidisciplinar y se engloba dentro de las prioridades del Plan Nacional de I+D+I. El objetivo general del proyecto es el estudio molecular de nuevos antimicrobianos (antibacterianos y antifúngicos) sintéticos diseñados a partir de estructuras privilegiadas de origen marino. El medio marino es una fuente de productos naturales extremadamente potentes que poseen actividades biológicas significativas. Prestaremos especial atención a determinar los posibles mecanismos de acción e interacción con los componentes celulares. Aprovecharemos el apoyo de la síntesis química para identificar los fragmentos de las moléculas implicados en la actividad, que hasta el momento sigue siendo una incógnita. Con ello pretendemos realizar estudios de relación estructura-‐actividad para diseñar productos de mejor perfil de actividad y eficacia, con especial énfasis en la selección de agentes activos contra las principales cepas involucradas en las infecciones nosocomiales. En el estudio de la actividad biológica se emplearán, siempre que sea posible, métodos de evalua-‐ción masiva y de uso general. Los métodos han sido desarrollados por los miembros del Grupo Solicitante durante sus trayectorias profesionales. Además, se aplicarán métodos de genómica, proteómica y farmacogenómica descritos ampliamente en la literatura científica. Estos nos permitirán evaluar el efecto de los productos en estudio sobre diversos aspectos de la biología celular de las bacterias y hongos en estudio. Los resultados obtenidos serán el punto de partida para desarrollar nuevos proyectos de investiga-‐ción y redactar artículos científicos para difundir nuestro trabajo. Los estudios mecanísticos pueden sentar las bases para el establecimiento de nuevas estrategias terapéuticas. Además, esperamos contribuir al conocimiento de las bases moleculares de las infecciones nosocomiales. Summary This proposal relates to a multidisciplinary project that falls within the priorities of the National Plan for Biomedicine. The main objective is the study of new synthetic antimicrobial (antibacterial and antifungal) agents designed from priviledged structures of marine origin. The marine environment is a source of ex-‐tremely potent natural products that have demonstrated significant biological activities.

We will focus on the elucidation of possible mechanisms of action and the interaction with cell com-‐ponents. With the aid of organic chemistry we will identify molecular fragments involved in activity, which remain unknown. With the obtained results we will perform structure-‐activity relationship studies to design products with better activity and efficacy profiles, with special interest in the selection of agents that are active against strains involved in nosocomial infections. In the assessment of the biological activity we will select when possible High Throughput Screening methods. The methods have been developed by the members of the group during their personal career and have been implemented at the host centers. We will also apply genomic, proteomic and pharmacogenomic methods described in the scientific literature. The methods will allow us to study the effect of the products on diverse items of the molecular biología of bacteria and fungi. The results will be the seed to start new research projects. Moreover, we plan to write a series of sci-‐entific papers in order to communicate the results of our research. The result of the mechanistic studies might establish new therapeutic strategies. Furthermore, we expect to increase the current knowledge in the molecular basis of nosocomial infections. The Spanish ion channel initiative Resúmen Los canales iónicos son proteínas de membrana que realizan un papel clave en todas las normas de vida, presentando una estructura altamente conservada de bacterias a humanos. La actividad de los canales iónicos ha sido reconocida como un soporte importante de la homeostasis celular y el mantenimiento de la salud por más de medio siglo. Debido a su contribución a la fisiopatogenia de varias enfermedades, estas proteínas han sido el blanco de diversos fármacos, desde antiepilépticos a analgésicos. Es más, la disfunción de los canales iónicos es por si misma la causa de muchas dolencias, acuñadas con el nombre de canalopatías. Más de 40 canalopatías asociadas a mutaciones genéticas de canales iónicos han sido descritas. A pesar de la intensa investigación en este excitante campo, existen todavía un elevado número de preguntas que carecen de respuesta, como son: i) la composición y papel de los complejos de señalización de canales iónicos en salud y enfermedad; iii) la estructura atómica de las subunidades que forman el poro de los canales y de sus complejos proteicos; iii) el conocimiento de las bases moleculares, estructurales y celulares de muchas canalopatías; y, iv) el descubrimiento de moduladores específicos que puedan ser útiles como herramientas farmacológicas y/o candidatos para fármacos. El estudio de estas complejas cuestiones requiere el establecimiento de un programa integrado de investigación pluridisciplinar en canales iónicos que aúne las metodologías necesarias y las coordine para cubrir exitosamente estas lagunas de nuestro conocimiento. Este es precisamente el objetivo primordial de IECI, convertirse en una referencia en investigación y formación que reúna todos los aspectos relacionados con los canales iónicos, desde genes, proteínas, células,

sistemas y organismos hasta comportamiento, desde ciencia básica a la clínica. Por tanto, IECI persigue la innovación mediante la integración. Los objetivos principales de IECI son: i) coordinar y facilitar programas de innovación científica que creen un centro virtual de excelencia en investigación integrada en canales iónicos; ii) formar a jóvenes investigadores en un amplio abanico de metodologías y técnicas en canales iónicos, incluyendo función, estructura, fisiopatología y farmacología; iii) estimular colaboraciones aportando los recursos, personal y equipamiento para proyectos conjuntos; iv) atraer nuevos grupos de investigación al campo de los canales iónicos; y, v) promover y fortalecer la interacción entre ciencia básica y clínica y la industria. Esta iniciativa integrada de investigación pluridisciplinar en canales iónicos está organizada en cuatro grupos/nodos basados en conocimiento que están altamente interrelacionados, a saber, función, estructura, fisiopatología y farmacología. Para conseguir sus objetivos, IECI reúne a un equipo multidisciplinar compuesto de fisiólogos, bioquímicos, biólogos moleculares y estructurales, biofísicos, farmacólogos, y químicos. Además, IECI no se crea como un consorcio cerrado sino que pretende ser una iniciativa abierta a todos aquellos grupos académicos, clínicos e industriales que Consolider 2008 deseen adherirse al programa. La aportación de IECI será la generación de conocimiento innovador en el campo de los canales iónicos que resultará en: i) tecnología novedosa transferible; ii) estrategias terapéuticas originales para el tratamiento de enfermedades mediadas por disfunción de los canales iónicos; y, iii) un programa de formación integrado en canales iónicos para estudiantes pre-‐ y post-‐doctorales. Por tanto, IECI cumplirá los dos objetivos cardinales de un programa multidisciplinar e integrado, a saber, crear conocimiento y tecnología innovadora y transferible e impulsar un programa formativo integrado. Summary Ion channels are membrane proteins that play crucial roles in all forms of life and are highly conserved from bacteria to humans. The activity of ion channels has been recogized as a important to cellular homeostasis and health maintenance for more than half a century. Because of their contribution to the patophysiology of several human maladies, these proteins have been the target of diverse drugs, from antiepileptic to analgesics. Furthermore, ion cannel dysfunction is itself the cause of human diseases, a pathological condition coined by the term channelopathy. More than 40 different channelopathies caused by genetic defects in ion channels have been reported. Despite the intense research in this exciting field, a plethora of issues remain poorly understood. These include: i) the composition, structure and role of signalling channel complexes in health and disease; ii) the atomic structure of the pore forming subunits and the channel complexes; iii) the knowledge on the molecular and cellular basis of channelopathies, as well as the implication of ion channels in the aetiology of other human diseases; iv) the discovery of more selective channel modulators that could be useful as pharmacological tools or drug leads. Addressing these key and complex questions requires an integrative and pluridisciplinary ion channel program that brings together all of the required complementary skills and coordinates them in order to successfully fill these gaps in our knowledge. This is precisely the main objective of SICI: to become a reference research and training programme that embraces all aspects of ion channels from genes and proteins, cells, systems and organisms to behaviour, and from basic research to the clinic. Thus, SICI pursues innovation through integration. The main objectives of SICI are: i) to coordinate and facilitate innovative research programmes that build on a virtual centre of excellence in integrative ion channel research; ii) to educate talented young scientists in a range of multidisciplinary methodologies and technologies in integrative ion channel research including channel function, structure, physiopathology and pharmacology; iii) to foster collaborative efforts by providing core resources, personnel and equipment for concerted research; iv) to bring new research groups and technologies to the field of integrative ion channel research; and v) to promote and strengthen the links between basic science, clinic and industry. This integrative ion channel research initiative is functionally organized into four distinct but highly interrelated, knowledge-‐based working groups or nodes encompassing channel function, structure, physiopathology and pharmacology. To accomplish this goal, SICI assembles a multidisciplinary team comprised of physiologists, biochemists, biophysicists, structural biologists, chemists and pharmacologists. SICI is not envisioned as a restricted consortium but rather Consolider 2008 seeks to be an open program for academic, clinical and industrial partners that may wish to join the initiative at a later stage. Cultivos de dinoflagelados marinos con interés alimentario, farmacológico y medioambiental Resumen Los dinoflagelados marinos son microalgas productoras de una gran variedad de biotoxinas. El crecimiento incontrolado de especies tóxicas de dinoflagelados (mareas rojas) en los ecosistemas acuaÅLticos de las costas andaluzas y de Espana, en general, genera un tremendo impacto en sectores productivos tales como el Turismo y la Industria extractiva del mar, ya que el consumo de crustáceos y moluscos contaminados con estas toxinas pueden ocasionar problemas de salud y cierre de caladeros. Muchas de estas toxinas son potentes sustancias bioactivas y presentan estructuras no encontradas en otros organismos marinos o terrestres, por lo que, en general, poseen mecanismos de acción novedosos. Ademaás estos microorganismos sintetizan una gran variedad de metabolitos de gran valor terapéutico. Esta situación ha generado una gran demanda de estas sustancias bioactivas en diferentes campos: control de calidad de alimentos marinos, toxicologáa, investigación química y médica, etc. Sin embargo, son inaccesibles en cantidad suficiente, limitando extraordinariamente su potencial. Muy pocas biotoxinas están disponibles a nivel comercial, ya las pocas que lo estaÅLn tienen precios desorbitados. La síntesis química de estas moléculas es muy compleja (cuando es posible) y extraordinariamente cara. La piedra angular de este problema radica en la posibilidad de cultivar masivamente esos microorganismos en condiciones controladas y reproducibles en el tiempo. Estos microorganismos parecen ser muy sensibles, entre otros factores, a las fuerzas de corte mecánicas e hidrodinámicas y al burbujeo, originando nuevos retos en la ingeniería de los fotobiorreactores. En este proyecto, esta escasez de conocimiento se pretende abordar aplicando y adaptando estrategias propias de la Ingeniería de Bioprocesos para intentar conseguir cultivos estables y de mediana escala desde el punto de vista del uso de los dinoflagelados marinos como factoriÅLas celulares de una gran

diversidad de compuestos bioactivos. Por tanto, teniendo en cuenta el estado actual del conocimiento científico sobre estos microorganismos y nuestra experiencia previa con ellos, se plantea como objetivo de este proyecto el establecer las bases de un bioproceso para obtener cultivos externos, a escala piloto, de dinoflagelados productores de biotoxinas y sustancias bioactivas en general. Se Utilizarán cepas de los géneros Protoceratium y Prorocentrum, productoras de yessotoxinas y acido okadaico, respectivamente (entre otras sustancias de interés). Las especies a utilizar han sido aisladas en las costas mediterráneas. Puesto que el principal problema es el desarrollo de cultivos estables a media-‐gran escala de dinoflagelados tóxicos, la resolución del mismo será el objetivo principal de este proyecto. Intentaremos conseguir lo que ya, a escala media, pero nada desdenable, hemos logrado utilizando luz artificial, con luz natural en cultivos externos. Se trabajará en estrategias para mitigar los efectos negativos de la turbulencia y el burbujeo en biorreactores, estudiando los principales mecanismos de dano celular. Paralelamente se trabajará en la mejora del medio de cultivo, en la optimización de las condiciones de crecimiento y modo de operacioÅLn de los sistemas de cultivo. Se intentará encontrar una estrategia de iluminación óptima y, con la información obtenida en esta tarea, se abordarán tareas de escalado de los cultivos en condiciones externas. Estrategia multidisciplinar en la terapia del cáncer: papel indiscutible de nuevas entidades químicas basadas en productos naturales (vegetal y marino) Resumen Las estructuras privilegiadas basadas en productos naturales constituyen un excelente punto de partida en el descubrimiento de nuevas entidades químicas con potencial anticancerígeno, ya que han sido seleccionadas por la naturaleza durante la evolución dada su capacidad para interactuar con una amplia variedad de proteínas y dianas biológicas. Además, la gran ventaja de los PN es “la novedad de sus estructuras, que difícilmente podrían ser el resultado de un razonamiento humano por creativo que fuese.” Las microalgas (incluyendo cianobacterias) constituyen un grupo muy heterogéneo de organismo con una amplia diversidad biológica. Esto les permite adaptarse prácticamente a todos los ecosistemas terrestres y acuáticos, lo que se traduce en una diversa maquinaría metabólica capaz de producir una amplia variedad de metabolitos de gran interés farmacéutico y biotecnológico. Por otro lado, son organismos cultivables y escalables en grandes volúmenes, no dependiendo de fuentes naturales, lo que los hacen interesantes para su posible aplicación industrial. Este proyecto permitirá aunar esfuerzos con la participación de dos entidades el IUBO (ULL) y el CBN (ULPG) con gran experiencia en la química de los productos naturales. Ello permitirá ampliar la diversidad estructural en la búsqueda nuevos agentes anticancerígenos. Por otra parte, la estrategia múltiple propuesta, actuando a nivel de prevención (agentes quimiopreventivos), quimioterápicos (agentes citotóxicos) e incluso revertidores de la multirresistencia a fármacos, permitirá abarcar diferentes dianas en la terapia del cáncer y por consiguiente las posibilidades de éxito. Estructuras privilegiadas obtenidas de fuentes vegetales en la prevención y tratamiento del cáncer
Resumen El cáncer constituye la segunda causa de mortalidad en los países desarrollados y el aumento de su incidencia explica el interés médico y social por la misma. Tanto los factores genéticos como medioambientales contribuyen al incremento en la morbilidad y mortalidad de distintos tipos de cáncer y entre estos factores se encuentran cambios en el estado redox celular como consecuencia de la sobreproducción de agentes oxidantes inducidos por mecanismos endógenos y exógenos que contribuyen al desarrollo del cáncer. Los vegetales son fuente de nuevos y más eficientes antioxidantes y citostáiticos a los que la farmacología puede recurrir para por un lado neutralizar los radicales libres producidos por las células y por otro a la antiproliferación de las células cancerosas. Por ello, es objetivo del presente proyecto estudiar el potencial antioxidante y antiproliferativo, de especies vegetales de Canarias e Iberoamerica que puedan resultar eficaces en la prevención y tratamiento del cáncer y así poder llegar a identificar y aislar estructuras privilegiadas con efectos terapéuticos mayores que las conocidas, o que operen por mecanismos novedosos, hecho que incrementaría el valor económico de tales plantas. Las sustancias de interés serán sometidas al estudio de su mecanismo de acción y a transformaciones químicas, lo que nos permitirá determinar los requerimientos estructurales para la expresión de la actividad y plantear el diseño racional de fármacos más potentes menos tóxicos y más selectivos. The iron(III) is able to catalyze this process with unactivated olefins. This cyclization was used as the key step in the shortest total synthesis of (+)-‐isolaurepan.

Their stereostructures have been elucidated on the basis of spectroscopic analysis, including 1D and 2D NMR techniques (COSY, ROESY, HSQC and HMBC), and spectrometric methods. The natural compounds and derivatives 13e15 have been tested for their potential as inhibitors of human immunodeficiency virus type 1 replication. Five compounds from this series displayed potent antiviral activity with IC50s in the micromolar range (1, 3, 4, 7 and 8) being 1 and 8 the most active compounds. The target of these compounds was different from antiretroviral drugs currently licensed as they act as inhibitors of enhancer-‐dependent transcription. The structureeactivity relationships were established based on the regiosubstitution and oxidation degree of the triterpene scaffold, revealing that these aspects were able to modulate the selectivity and intensity of HIV inhibition.

Withania aristata. Their structures were elucidated on the basis of spectroscopic analysis, including 1D and 2D NMR techniques. Semisynthesis of the minority metabolites 7 and 15 from compounds 6 and 9, respectively, as starting material, was performed. The isolated compounds as well as three derivatives (7a, 9a and 9b) of withaferin A were evaluated for cytotoxicity against HeLa (carcinoma of the cervix), A-‐ 549 (lung carcinoma) and MCF-‐7 (breast adenocarcinoma) human cancer cell lines, and against normal Vero cells (African green monkey kidney). Five compounds from this series (8, 9a, 9b, 11 and 13) exhibited potent antiproliferative effects on the tumor cells, even higher than the well known anticancer agent, withaferin A (9). Phosphatidylserine externalization, chromatin condensation, and caspase-‐3 activation clearly indicated apoptosis as a mechanism of action. The structureeactivity relationship revealed valuable information on the pharmacophore for withanolide-‐type compounds.

Type 2 diabetes mellitus (T2DM) is a rapidly expanding public epidemic and frequently results in severe vascular complications. In an attempt to find anti-‐diabetic agents, we report herein on the isolation, structural elucidation and bioactivity of nine friedelane-‐type triterpenes (1–9) and twenty two known ones (10–31) from the root barks of Celastrus vulcanicola and Maytenus jelskii. Their structures were elucidated on the basis of spectroscopic analysis, including 1D and 2D NMR techniques. Two compounds from this series (1 and 3) exhibited increased insulin-‐mediated signalling, which suggests these friedelane triterpenes have potential therapeutic use in insulin resistant states.

Three new triterpenoids, including two rare D:B-‐friedobaccharanes (leonatriol, 1 and leonatriolone, 2) and a 2,3-‐seco-‐2,24-‐epoxy-‐3,24-‐olide-‐D:A-‐friedooleanane (cassinolide, 3) were isolated from the root bark of Cassine xylocarpa and Celastrus vulcanicola, collected in El Salvador. Their stereostructures were elucidated on the basis of spectroscopic analyses, mainly 1D and 2D NMR techniques, spectrometric analyses, and comparison with data reported in the literature. The absolute configuration of 1 and 2 were determined by the application of the Riguera ester procedure and biogenetic considerations. The posible biogenetic pathway for cassinolide (3) is also discussed.

In this work, the use of the ionic liquid (IL) 1,3-‐dipentylimidazolium hexafluorophosphate ([PPIm][PF6]) as an alternative extractant for IL dispersive liquid–liquid microextraction (IL-‐DLLME) of a group of pesticides and metabolites (2-‐aminobenzimidazole, carbendazim/ benomyl, thiabendazole, fuberidazole, carbaryl, 1-‐naphthol, and triazophos) from soils is described. After performing an initial ultrasound-‐assisted extraction (USE), the IL-‐DLLME procedure was applied for the extraction of these organic analytes from soil extracts. Separation and quantification was achieved by high-‐performance liquid chromatography (HPLC) with fluorescence detection (FD). Calibration, precision, and accuracy of the described USE-‐IL-‐DLLME-‐HPLC-‐FD method using [PPIm][PF6] as an alternative extractant was evaluated with two soils of different physicochemical properties. Accuracy percentages were in the range 93–118% with RSD values below 20%. A comparison of the performance of [PPIm][PF6] versus that of the so-‐common 1-‐hexyl-‐3-‐methylimidazolium hexafluorophosphate ([HMIm][PF6]) was accomplished. Results indicate a comparable extraction efficiency with both ILs, being slightly higher with [HMIm][PF6] for the metabolite 2-‐aminobenzimidazole, and slightly higher with [PPIm][PF6] for triazophos. In all cases, LODs were in the low ng/g range (0.02–14.2 ng/g for [HMIm][PF6] and 0.02–60.5 ng/g for [PPIm][PF6]). As a result, the current work constitutes a starting point for the use of the IL [PPIm][PF6] for further analytical approaches.

The Lewis acid mediated reaction of acyclic vinyl allenes and imines, yielding tetrahydropyridines has been carried out. Only one cycloadduct was observed in each case, and thus, the reaction progressed in all cases with total regio-‐, face-‐ and endo/exo selectivities. The cycloadducts were transformed into polysubstituted pyridines, including bipyridines, by catalytic transfer hydrogenation using cyclohexene as hydrogen donor in good yield. The reaction was extended to the aromatization of the bicyclic cycloadducts prepared in previous works from the reaction of semicyclic vinyl allenes and imines, yielding tetrahydropyridines

A series of alkyl a/b-‐(1?6)-‐diglucopyranosides 1_12 were synthesized and assessed for cytotoxicity against HL-‐60, U937, Molt-‐3 and MCF-‐7 cancer cell lines. The menthyl derivatives displayed strong cytotoxic properties showing IC50 values between 6 and 16 lM. Furthermore, we demonstrated that the selected synthetic (+)-‐menthyl b-‐(1?6)-‐diglucopyranoside 5 induces apoptotic cell death in human leukemia cells through a mechanism that involves activation of multiple caspases. Cell death was completely prevented by the non-‐specific caspase inhibitor z-‐VAD-‐fmk and found to be associated with the release of cytochrome c, an increase in the expression of Bax levels and a decrease in the generation of reactive oxygen species.

A series of carbasugars were prepared and their conformational properties studied by means of NMR spectroscopy. The results were compared to those previously found for O-‐, S-‐, and C-‐b-‐glycoside analogs. While the rotational populations of the hydroxymethyl group in O-‐, S-‐, and C-‐glycosides are known to depend on the structural nature of their aglycon, in carbasugars it proved to be independent of the pseudo-‐aglycon. This result confirms that endocyclic oxygen is necessary for the observed relationship between the structure of the aglycon and the rotational populations of the hydroxymethyl group, and indicates that the stereoelectronic exo-‐anomeric effect is mainly responsible for such conformational dependence.

A new family of heterometallic compounds 3–6 containing ferrocenyl and platinum(II) centers has been synthesized by reaction of 1-‐b-‐aminoethylferrocene (1) and 1,1¢-‐bis(b-‐aminoethyl)ferrocene (2) with Pt(II) precursors. Using K2[PtCl4] as the Pt(II) source, the cis-‐square-‐planar neutral compounds [Fe{h5-‐C5H4(CH2)2NH2}2PtCl2] (3) and [{Fe(h5-‐C5H4(CH2)2NH2)(h5-‐C5H5)}2PtCl2] (5) were obtained. Reaction of cis-‐[PtCl2(dmso)2] with 1 and 2 resulted in the displacement of dmso and chloride ligands from the platinum coordination sphere, affording the cationic and neutral compounds [Fe{h5-‐C5H4(CH2)2NH2}2Pt(dmso)Cl]Cl (4) and [Fe(h5-‐C5H4(CH2)2NH2)(h5-‐C5H5)Pt(dmso)Cl2] (6). Compounds 3–6 were thoroughly characterized using multinuclear (1H, 13C, 195Pt) NMR, IR spectroscopy, ESI mass spectrometry and elemental analysis. Single-‐crystal X-‐ray analysis of heterometallic 6 confirmed the cis geometry of the molecule and revealed that the platinum atom is held in a perfect square-‐planar geometry. The electrochemical behaviour of the heterometallic compounds 3–6, which has been examined by cyclic (CV) and square wave (SWV) voltammetries in dichloromethane and dmso solution, is characterized by the reversible one-‐electron oxidation of the ferrocene moieties. The results of the biological activity studies revealed that the organometallic complex 5 is active against all cell lines with GI50 values in the range 1.7–2.3 mM. When compared to the standard anticancer drug cisplatin, heterotrimetallic 5, possessing two aminoethylferrocenyl units coordinated to the Pt(II) center, showed a greater activity profile in the colon cancer cell line. Cell cycle studies revealed that the new mixed compound exhibits a mechanism of action different to cisplatin.

In this study, we describe the synthesis of a series of α-‐ and β-‐lapachone containing hydroxyl or methoxyl groups on the benzene ring, by means of the selective acid promoted cyclization of the appropriate lapachol analog. The evaluation of the antiproliferative activity in human solid tumor cell lines provided 7-‐hydroxy-‐β -‐lapachone as lead with enhanced activity over the parent drug β-‐lapachone. Cell cycle studies, protein expression experiments, and reactive oxygen species analysis revealed that, similarly to β-‐lapachone, ROS formation and DNA damage are critical factors in the cellular toxicity of 7-‐ hydroxy-‐β-‐lapachone.

Background: Despite our increased understanding of the mechanisms involved in acute lung injury (ALI) and the acute respiratory distress syndrome (ARDS), there is no specific pharmacological treatment of proven benefit. We used a novel screening methodology to examine potential anti-‐inflammatory effects of a small structure-‐focused library of synthetic carbamate and urea derivatives in a well established cell model of lipopolysaccharide (LPS)-‐induced ALI/ARDS. Methodology/Principal Findings: After a pilot study to develop an in vitro LPS-‐induced airway epithelial cell injury model, a library of synthetic carbamate and urea derivates was screened against representative panels of human solid tumor cell lines and bacterial and fungal strains. Molecules that were non-‐cytotoxic and were inactive in terms of antiproliferative and antimicrobial activities were selected to study the effects on LPS-‐induced inflammatory response in an in vitro cell culture model using A549 human alveolar and BEAS-‐2B human bronchial cells. These cells were exposed for 18 h to LPS obtained from Escherichia coli, either alone or in combination with the test compounds. The LPS antagonists rhein and emodin were used as reference compounds. The most active compound (CKT0103) was selected as the lead compound and the impact of CKT0103 on pro-‐inflammatory IL-‐6 and IL-‐8 cytokine levels, expression of toll-‐like receptor-‐4 (TLR4) and nuclear factor kappa B inhibitor alpha (IkBa) was measured. CKT0103 significantly inhibited the synthesis and release of IL-‐6 and IL-‐8 induced by LPS. This suppression was associated with inhibition of TLR4 up-‐regulation and IkBa down-‐regulation. Immunocytochemical staining for TLR4 and IkBa supported these findings. Conclusions/Significance: Using a novel screening methodology, we identified a compound – CKT0103 – with potent antiinflammatory effects. These findings suggest that CKT0103 is a potential target for the treatment of the acute phase of sepsis and sepsis-‐induced ALI/ARDS.

Three withanolides were isolated from the aerial parts of Jaborosa reflexa Phil. Jaborosa cabrerae Barboza yielded five sativolide withanolides (including jaborosalactones R, S, 38, and 39) and two trechonolide withanolides epimeric at C-‐23 (trechonolide A and jaborosalactone 32). In addition, five derivatives were obtained by chemical derivatization of jaborosalactone 38, and all compounds were fully characterized by 1D and 2D NMR spectroscopic studies. The in vitro antiproliferative activities of the major natural withanolides and the semisynthetic derivatives were examined against HBL-‐100, HeLa, SW1573, T-‐47D, and WiDr human solid tumor cancer cell lines. Some chemotaxonomic considerations are discussed.

DTA0100 is a new catalytic inhibitor of the human DNA topoisomerase IIα that induces G2/M phase cell cycle arrest in human solid tumor cells lines from various malignancies. In our study, we investigated the effectiveness of the combined treatment of ionizing radiation with DTA0100 on the survival of three representative human solid tumor cell lines: HeLa (cervix), WiDr (colon) and SW1573 (non-‐small cell lung cancer). The concomitant treatment of DTA0100 and irradiation showed a synergistic and antagonistic effect in the three cell lines tested. A synergistic cytotoxic effect of the combination of DTA0100 and radiation was confirmed by the median drug effect analysis method. It was found that in those protocols where the drug was administered after radiation the most synergistic effect was achieved. Our study constitutes the first in vitro evidence for synergistic effects between DTA0100 and radiation. This combination therapy might thus be expected to be more effective than either treatment alone in patients with cervical, colon and non-‐small cell lung cancer cells.

Six diiodido−diamine platinum(II) complexes, either cis or trans configured, were prepared, differing only in the nature of the amine ligand (isopropylamine, dimethylamine, or methylamine), and their antiproliferative properties were evaluated against a panel of human tumor cell lines. Both series of complexes manifested pronounced cytotoxic effects, with the trans isomers being, generally, more effective than their cis counterparts. Cell cycle analysis revealed different modes of action for these new Pt(II) complexes with respect to cisplatin. The reactivity of these platinum compounds with a number of biomolecules, including cytochrome c, two sulfur containing modified amino acids, 9-‐ethylguanine, and a single strand oligonucleotide, was analyzed in depth by mass spectrometry and NMR spectroscopy. Interestingly, significant differences in the reactivity of the investigated compounds toward the various model biomolecules were observed: in particular we observed that trans complexes preferentially release their iodide ligands upon biomolecule binding, while the cis isomers may release the amine ligands with retention of iodides. Such differences in reactivity may have important mechanistic implications and a relevant impact on the respective pharmacological profiles.

The best activity profiles were obtained for the derivatives bearing more lipophilic esters (GI50 3.1-‐21 μM).

Objectives. To evaluate the in vitro leishmanicidal activity of nine flavonoid derivatives from Delphinium staphisagria against L. infantum and L. braziliensis. Design and Methods. The in vitro activity of compounds 1–9 was assayed on extracelular promastigote and axenic amastigote forms and on intracellular amastigote forms of the parasites. Infectivity and cytotoxicity tests were carried on J774.2 macrophage cells using Glucantime as the reference drug. The mechanisms of action were analysed performing metabolite excretion and transmission electronic microscope ultrastructural alteration studies. Results.Nine flavonoids showed leishmanicidal activity against promastigote as well as amastigote forms of Leishmania infantum and L. braziliensis. These compounds were nontoxic to mammalian cells and were effective at similar concentrations up to or lower than that of the reference drug (Glucantime). The results showed that 2 -‐acetylpetiolaroside (compound 8) was clearly the most active. Conclusion. Thisstudy has demonstrated that flavonoid derivatives are active against L. infantum and L. braziliensis.

To achieve a more exact understanding of the water transport in natural channels, a series of non-‐natural structures have been developed. They have been studied by far-‐infrared spectroscopy, solid-‐state nuclear magnetic resonance, differential scanning calorimetry, thermogravimetric analysis, and variable-‐temperature powder X-‐ray diffraction to examine the behavior of water at the molecular level. Water in these predominantly nonpolar pores can be metastable, with filling and emptying occurring upon changes in solvent conditions. The water contained in these pores exhibits a dynamics that might be controlled, since it depends on the structural features of the monomers that form the pore “skeleton”. We have observed changes in the pore diameter depending on the selected isomer. This provokes at a given temperature differences in the arrangement and dynamics of the contained water. The water dynamics increases with both temperature and pore diameter in a process that is reversible over a temperature range specific for each structure. Beyond this particular temperatura threshold, the pore water can be irreversibly evacuated, and at this point a decrease of the dynamics is observed. The slower dynamics of the remaining water in partially evacuated pores is probably due to the increased interaction with the inner-‐pore surface owing to a concomitant narrowing of the pore. These findings not only highlight the need for the presence of freely moving water inside the pore to sustain its permeability by water, but also point to the decrease in the dynamics of the remaining water in partially evacuated pores.

The dinoflagellate Prorocentrum belizeanum is responsible for the production of several toxins involved in the red tide phenomenon known as Diarrhetic Shellfish Poisoning (DSP). In this paper we report on the biosynthetic origin of an okadaic acid water-‐soluble ester derivative, DTX5c, on the basis of the spectroscopical analysis of 13C enriched samples obtained by addition of labelled sodium [l-‐13C], [2-‐13C] acetate to artificial cultures of this dinoflagellate.

Two new cytotoxic oxasqualenoids, saiyacenols A and B, were isolated from the red alga Laurencia viridis and their structures elucidated by spectroscopic methods and chemical correlation with the well-‐known compound dehydrothyrsiferol. The new compounds share an important part of their structure with thyrsiferol and aplysiols B and C. Isolation of the saiyacenols concludes the controversy about the configuration of aplysiols, correcting the structure of aplysiol B. Our results illustrate the current limitations of NMR J-‐based methods when approaching quaternary carbons and demand a reappraisal of rational biogenetic proposals in structural elucidation.

Chemical study of marine organisms has revealed them as a rich source of natural products with unique structural characteristics and outstanding biological activities. Marine macrolides are a unique group of natural products that frequently include a highly oxygenated polyene backbone and a macrocyclic lactone as a conformational constraint. Many of them have shown unparalleled cell growth antiproliferative properties, making them valuable molecular probes for the discovery of new biochemical pathways or as promising lead compounds in the development route to new antitumor chemotherapeutic agents. This bibliographic review has been focussed on marine macrolides with strong cytotoxic activity and potential in cancer research and therapy, as well as those macrolides either in the market or currently in clinical trials and/or preclinical development.

Phytochemistry, 2012, 81, 60-‐70 Biotransformation of ent-‐kaur-‐16-‐ene and ent-‐trachylobane 7b-‐acetoxy derivatives by the fungus Gibberella fujikuroi (Fusarium fujikuroi) Braulio M. Fraga, Carlo Bressa, Victoria González-‐Vallejo, Pedro González, Ricardo Guillermo
Candol A (7β-‐hydroxy-‐ent-‐kaur-‐16-‐ene) (6) is efficiently transformed by Gibberella fujikuroi into the gibberellin plant hormones. In this work, the biotransformation of its acetate by this fungus has led to the formation of 7β-‐acetoxy-‐ent-‐kaur-‐16-‐en-‐19-‐oic acid (3), whose corresponding alcohol is a short-‐lived intermediate in the biosynthesis of gibberellins and seco-‐ring ent-‐kaurenoids in this fungus. Further biotransformation of this compound led to the hydroxylation of the 3b-‐positions to give 7b-‐acetoxy-‐3b-‐hydroxy-‐ent-‐kaur-‐16-‐en-‐19-‐oic acid (14), followed by a 2β-‐ or 18-‐hydroxylation of this metabolite. The incubation of epicandicandiol 7β-‐monoacetate (7β-‐acetoxy-‐18-‐hydroxy-‐ent-‐kaur-‐16-‐ene) (10) producesalso the 19-‐hydroxylation to form the 18,19 diol (20), which is oxidized to give the corresponding C-‐18 or C-‐19 acids. These results indicated that the presence of a 7β-‐acetoxy group does not inhibit the fungal oxidation of C-‐19 in 7β-‐acetoxy-‐ent-‐kaur-‐16-‐ene, but avoids the ring B contraction that leads to the gibberellins and the 6β-‐hydroxylation necessary for the formation of seco-‐ring B ent-‐kaurenoids. The biotransformation of 7b-‐acetoxy-‐ent-‐trachylobane (trachinol acetate) (27) only led to the formation of 7β-‐acetoxy-‐18-‐hydroxy-‐ent-‐trachylobane (33).

The biotransformation of 13a,17-‐dihydroxystemodane (3) with the fungus Cephalosporium aphidicola afforded 13a,17,18-‐trihydroxystemodane (4), 3b,13a,17-‐trihydroxystemodane (5), 13a,17-‐dihydroxy-‐stemodan-‐18-‐oic acid (6), 3b,11β,13a,17-‐tetrahydroxystemodane (7), 11β,13a,17,18-‐tetrahydroxystemodane (8) and 3b,13a,17,18-‐tetrahydroxystemodane (9). The hydroxylation at C-‐18 of the substrate points to a biosynthetically-‐directed transformation, because aphidicolin (2) is hydroxylated at this carbon. However, the C-‐3(b) and C-‐11(b) hydroxylations seem to indicate a xenobiotic biotransformation.

Thirty one derivatives were prepared from the natural alkaloids haemanthamine (1), haemanthidine (2) and 11-‐hydroxyvittatine (3). They were evaluated for their in vitro antimalarial activity against chloro-‐ quine-‐sensitive strains of Plasmodium falciparum and some structure–activity relationships were out-‐ lined. For haemanthamine derivatives having a methoxy group at C-‐3, the presence of a free hydroxyl group at C-‐11 is important for the activity. The double bond at C-‐1–C-‐2 plays also an important role to achieve good inhibitory activity. Compound 35 with two nicotinate groups at C-‐3 and at C-‐11 was the most active compound with a IC50 = 0.8 ± 0.06 lM.

Labdane derivatives obtained from the diterpenoid labdanediol suppressed NO and PGE2 production in LPSstimulated RAW 264.7 macrophages. However, mechanisms involved in these inhibitory effects are not elucidated. In this study, we investigated the signaling pathways involved in the anti-‐inflammatory effects of labdanolic acid methyl ester (LAME) in peritoneal macrophages and examined its therapeutic effect in a mouse endotoxic shock model. LAME reduced the production of NO and PGE2 in LPS-‐activated macrophages. This effect involved the inhibition of NOS-‐2 and COX-‐2 gene expression, acting at the transcription level. Examination of the effects of the diterpene on NF-‐κB signaling showed that LAME inhibits the phosphorylation of IκBα and IκBβ, preventing their degradation and the nuclear translocation of the NF-‐κB p65 subunit. Moreover, inhibition of MAPK signaling was also observed. A further experiment revealed that LAME inhibited the phosphorylation of transforming growth factor-‐β (TGF-‐β)-‐activated kinase 1 (TAK1), an upstream signaling molecule required for IKK and mitogen-‐activated protein kinases (MAPKs) activation. Inflammatory cytokines such as IL-‐6, TNF-‐α and IP-‐10 were downregulated in the presence of this compound after stimulation with LPS. Additionally, LAME also improved survival in a mouse model of endotoxemia and reduced the circulatory levels of cytokines (IL-‐6, TNF-‐α). In conclusion, these results indicate that labdane diterpene LAME significantly attenuates the pro-‐inflammatory response induced by LPS both in vivo and in vitro.

Several disubstituted angular naphthoimidazoles have been synthesized by a new domino reaction from (1,4-‐dimethoxynaphthalen-‐ 2-‐yl)amine (1) and imines (obtained from 1 and aromatic aldehydes) in the presence of Sc(OTf)3. A plausible pathway for the reaction involves attack of the nucleophilic amine on the imine and further reaction of the resulting intermediate by intramolecular cyclization with loss of a methoxy group and subsequent oxidation to yield the final naphthoimidazole. These naphthoimidazoles were also obtained in lower yields from aromatic aldehydes, Sc(OTf)3 and 2 equiv. of 1 through an ABB_ domino process. Post-‐condensation transformations allowed direct access to several derivatives.

Secretory vesicles of chromaffin cells are acidic organelles that maintain an increasing pH gradient towards the cytosol (5.5 vs. 7.3) that is mediated by V-‐ATPase activity. This gradient is primarily responsable for the accumulation of large concentrations of amines and Ca2+, although the mechanisms mediating Ca2+ uptake and release from granules, and the physiological relevance of these processes, remain unclear. The presence of a vesicular matrix appears to create a bi-‐compartmentalised medium in which the major fractions of solutes, including catecholamines, nucleotides and Ca2+, are strongly associated with vesicle proteins, particularly chromogranins. This association appears to be favoured at acidic pH values. It has been demonstrated that disrupting the pH gradient of secretory vesicles reduces their rate of exocytosis and promotes the leakage of vesicular amines and Ca2+, dramatically increasing the movement of secretory vesicles and triggering exocytosis. In this short review, we will discuss the data available that highlights the importance of pH in regulating the association between chromogranins, vesicular amines and Ca2+. We will also address the potential role of vesicular Ca2+ in two major processes in secretorycells, vesicle movement and exocytosis.

Chromogranins A (CgA) and B (CgB) are the main soluble proteins of large dense-‐core secretory vesicles (LDCVs). Using CgA-‐ and CgB-‐knockout (KO) mice, we found that the absence of chromogranins A and B induces significant changes in catecholamine (CA) accumulation and the kinetics of exocytosis. By crossing these two knockout strains, we generated a viable and fertile double CgA/BKO mouse in which the catecholamine content in chromaffin LDCVs was halved, and the secretory response significantly reduced. Incubating cells with L-‐DOPA increased the vesicular CA content in wild-‐type (WT) but not in Cg-‐KO cells, which was not due to changes in amine transport, or in the synthesis or degradation of cytosolic amines. Electron microscopy revealed the presence of giant secretory vesicles exhibiting significant alterations, with little or no electrodense inner matrix. Proteomic analysis confirmed the absence of CgA and B, and revealed small changes in SgII in the LDCV-‐enriched fraction, as well as the overexpression of fibrinogen and other proteins. In summary, our findings indicate that the mechanisms responsible for vesicular accumulation of CA are saturated in Cgs-‐KO cells, in contrast to the ample capacity for further accumulation in WTcells. We conclude that Cgs contribute to a highly efficient system that directly mediates monoamine accumulation and exocytosis in LDCVs.

Faithful replication of DNA molecules by DNA polymerases is essential for genome integrity and correct transmission of genetic information in all living organisms. DNA polymerases have recently emerged as important cellular targets for chemical intervention in the development of anti-‐cancer agents. Herein we report additional synthesis of simplified bicyclic aglycones of iridoids and their biological activity against Taq DNA polymerase with the object to find out some of the likely molecular targets implicated in the biological activity showed for this kind of compounds. The compounds 14, 33 and 34 showed inhibitory activity against Taq DNA polymerase with IC50 values of 13.47, 17.65 and 18.31 μM, respectively. These results would allow proposing to DNA polymerases as the molecular targets implicated in this bioactivity and enhance the iridoid aglycones as leader molecule to develop new drugs for cancer therapy.

Cultured chromaffi n cells have been used for almost 40 years in the study of different cell functions using biochemical, electrophysiological, pharmacological, and toxicological approaches. Chromaffi n cells are essentially secretory cells that are used to model sympathetic neurons or neuroendocrine cells. In this chapter, we describe the most common methods currently used to isolate and culture chromaffi n cells from the animals used most commonly: cows, rats, and mice. We also provide some advice on the use of these cells in the laboratory.

Small peptides patterned after the N terminus of the synaptosomal protein of 25 kDa, a member of the protein complex implicated in Ca2_-‐dependent neuronal exocytosis, inhibit in vitro the release of neuromodulators involved in pain signaling, suggesting an in vivo analgesic activity. Here, we report that compound DD04107 (palmitoyl-‐EEMQRR-‐NH2), a 6-‐mer palmitoylated peptide that blocks the inflammatory recruitment of ion channels to the plasma membrane of nociceptors and the release of calcitonin gene-‐related peptide from primary sensory neurons, displays potent and long-‐lasting in vivo antihyperalgesia and antiallodynia in chronic models of inflammatory and neuropathic pain, such as the complete Freund’s adjuvant, osteosarcoma, chemotherapy, and diabetic neuropathic models. Subcutaneous administration of the peptide produced a dose-‐dependent antihyperalgesic and antiallodynic activity that lasted _24 h. The compound showed a systemic distribution, characterized by a bicompartmental pharmacokinetic profile. Safety pharmacology studies indicated that the peptide is largely devoid of side effects and substantiated that the in vivo activity is not caused by locomotor impairment. Therefore, DD04107 is a potent and long-‐lasting antinociceptive compound that displays a safe pharmacological profile. These findings support the notion that neuronal exocytosis of receptors and neuronal algogens pivotally contribute to chronic inflammatory and neuropathic pain and imply a central role of peptidergic nociceptor sensitization to the pathogenesis.

Chromogranins are the main soluble proteins in the large dense core secretory vesicles (LDCVs) found in aminergic neurons and chromaffin cells. We recently demonstrated that chromogranins A and B each regulate the concentration of adrenaline in chromaffin granules and its exocytosis. Here we have further studied the role played by these proteins by generating mice lacking both chromogranins. Surprisingly, these animals are both viable and fertile. Although chromogranins are thought to be essential for their biogenesis, LDCVs were evident in these mice. These vesicles do have a somewhat atypical appearance and larger size. Despite their increased size, single-‐cell amperometry recordings from chromaffin cells showed that the amine content in these vesicles is reduced by half. These data demonstrate that although chromogranins regulate the amine concentration in LDCVs, they are not completely essential, and other proteins unrelated to neurosecretion, such as fibrinogen, might compensate for their loss to ensure that vesicles are generated and the secretory pathway conserved.—Díaz-‐ Vera, J., Camacho, M., Machado, J. Galanthamine-‐type alkaloids produced by plants of the Amaryllidaceae family are potent acetylcholinesterase inhibitors. One of them, galanthamine, has been marketed as a hydrobromide salt for the treatment of Alzheimer’s disease. In the present work, gas chromatography with electron impact mass spectrometry (GC-‐EIMS) fragmentation of 12 reference compounds isolated from various amaryllidaceous plants and identified by spectroscopic methods (1D and 2D nuclear magnetic resonance, circular dichroism, high-‐resolution MS (HRMS) and EIMS) was studied by tandem mass spectrometry (GC-‐MS/MS) and accuratemassmeasurements (GC-‐HRMS). The studied compounds showed good peak shape and efficient GC separation with a GC-‐MS fragmentation pattern similar to that obtained by direct insertion probe. With the exception of galanthamine-‐N-‐oxide and N-‐formylnorgalanthamine, the galanthamine-‐type compounds showed abundant [M]+. and [M-‐H]+ ions. A typical fragmentation pattern was also observed, depending on the substituents of the skeleton. Based on the fragmentation pathways of reference compounds, three other galanthamine-‐type alkaloids, including 3-‐O-‐(2´-‐butenoyl)sanguinine, which possesses a previously unelucidated structure, were identified in Leucojum aestivum ssp. pulchelum, a species endemic to the Balearic islands. GC-‐MS can be successfully applied to Amaryllidaceae plant samples in the routine screening for potentially new or known bioactive molecules, chemotaxonomy, biodiversity and identification of impurities in pharmaceutical substances.

Lupane triterpenoids 2 and 5−12 and oleanene derivatives 13 and 14 were prepared from lupeol (1), betulin (3), and germanicol (4). They were tested for anti-‐HIV activity, and some structure−activity relationships were outlined. The 20-‐(S) absolute configuration of epoxylupenone (8) was assessed by comparison of the observed and DFTcalculated vibrational circular dichroism spectra. The CompareVOA algorithm was employed to support the C-‐20 configuration assignment. The 20,29 double bond in lupenone (2) and 3-‐epilupeol (15) was stereoselectively epoxidized to produce 20-‐(S)-‐8 and 20-‐(S)-‐16, respectively, an assignment in agreement with their X-‐ray diffraction structures.

ATP being probably the first neurotransmitter, it seems plausible that the history of secretion started from the fusion of an ATP-‐contained vesicle with the cell membrane. Newly created transmitters found purinergic vesicles already made. All vesicles accumulate H+: the pH, the electric gradient or both are required for concentrating most transmitters inside. The proton gradient is produced by the V-‐ATPase, which is also activated by ATP, another fact to think about. Neverthe-‐ less, although this commentary will not change any biological fact, we should consider that perhaps exocytosis of ATP is the pillar for understanding cell-‐to-‐cell communication.

The Lycopodium alkaloids are quinolizidine or pyridine and a-‐pyridone type alkaloids from the Lycopodium L., Huperzia Bernh., Lycopodiella Holub, and Phylloglosum Kunze genera, which have unique heterocyclic skeletons of C11N, C15N2, C16N, C16N2, C22N2, and C27N3. During recent years, this group of alkaloids has gained great interest due to their important pharmacological properties as well as for their complex molecular architectures.

Ten indole alkaloids were isolated from Geissospermum reticulatum, seven (1−7) from the leaves and three (8−10) from the bark. Seven were aspidospermatan-‐type alkaloids (1−3, 5−9), including four (5−8) with a 1-‐oxa-‐3-‐cyclopentene group in their molecule, which we named geissospermidine subtype. Compounds 1−3, 5−8, and 10 had not been reported previously as natural products, while 4 and 9 were the known alkaloids Odemethylaspidospermine and flavopereirine. Their structures were determined by spectroscopic techniques including 1D and 2D NMR experiments (COSY, NOESY, HSQC, HMBC). Additionally, X-‐ray crystallographic analyses of 1, 2, and 6 were performed. Antiparasitic activities of the ethanolic and alkaloidal extracts and of the pure alkaloids were tested against Trypanosoma cruzi and Leishmania infantum. In general, the extracts exhibited selective action and were more active against Leishmania than against Trypanosoma. Alkaloid 4 was also very active against L. infantum.

A novel supramolecular Cu(I) complex {Cu2I2L}n (L: propane-‐1,3-‐diyl bis-‐(4-‐pyridine-‐carboxylate)) (1) has been prepared by self-‐assembly reaction of CuI and the flexible ligand (L) using an acetonitrile and acetonitrile/water (4:1) and mixture respectively, in presence of an excess of KI. X-‐ray diffraction analyses revealed that (1) is one-‐dimensional coordination polymer with double-‐stranded staircase motif. The structure was also characterized by i.r. spectroscopy, and thermogravimetry. In the solid state at room temperature, the compound exhibits orange phosphorescence with emission maxima at 590 nm.

Eleven eremophilanolides, 1–3 and 6–13, and two eremophilanes, 24 and 25, were isolated from Senecio candidans and S. magellanicus from the Magallanes Region (Chile). Compounds 2, 3, 9, and 10 have not been previously reported as natural products. Their structures were established by NMR spectroscopic analysis and chemical transformations. The X-‐ray analysis of compounds 11, 13, and 17 were also performed. Different semisynthetic analogs from eremophilanolide 11 were generated to carry out a structure_activity relationship study. Their possible plant defensive role was tested against herbivorous insects (Spodoptera littoralis, Rhopalosiphum padi, and Myzus persicae) and plants (Lactuca sativa). Additionally, their effects on insect (Sf9) and mammalian (CHO) cell lines were tested.

The 2-‐amino-‐Nʹ′-‐phenylbenzohydrazide 2 is a key intermediate to obtain quinazolinones and benzotriazepines derivatives. The quinazolinone nucleous and its derivatives have been extensively studied because of their wide range of pharmacological activities, including antiviral, antibacterial, antifungal, antimalarial, anticancer, antihypertensive, diuretic, anticonvulsant and anti-‐inflammatory (Kamal et al., 2010). On the other hand, the benzotriazepinones have been described as efficient enzymatic inhibitors (Filippakopoulos et al., 2012; Spencer et al., 2008). We report herein on the synthesis and crystal structure of the title compound, a member of this important family of compounds.

The title compound, C16H14O5_2H2O [systematic name: 5-‐hydroxy-‐2-‐(4-‐hydroxyphenyl)-‐7-‐methoxychroman-‐4-‐one dihydrate], is a natural phytoalexin flavone isolated from the native chilean species Heliotropium taltalense and crystallizes with an organic molecule and two water molecules in the asymmetric unit.

The design of polymeric organic-‐inorganic materials with novel topologies and structural motifs is of current interest in the field of coordination chemistry, (Blake et al., 1999). This paper forms part of our continuing study of the synthesis, structural characterization and physical properties of coordination polymers (Brito et al., 2011). The title compound was isolated during attempts to synthesize a mixed-‐ligand coordination polymer by solvothermal reaction. Our initial model, with mixed ligands, was not considered because the compound crystallizes in the space group P1 with CuII located at a centre of symmetry so the model was changed to considering both ligands separately.
