III. Cystic fibrosis 9 points “Cystic fibrosis is one of the most common and best studied inherited ion channel disorders, which is caused by the lack of ion channels in glandular epithelial cells. On average, 1 out of every 2500 infants carries the recessive mutation in a homozygous form. Heterozygous individuals show no symptoms of the disease and they are mostly unaware that they are carriers. Although cystic fibrosis affects various organs, including the intestine, pancreas and reproductive tract, the respiratory tract usually shows the most severe symptoms. Victims of the disease produce thickened, sticky mucus, which is very hard to propel out of the airways. Affected individuals usually suffer from chronic lung inflammation. The gene responsible for cystic fibrosis (CF) was identified in 1989 and soon the base sequence of the gene and the order of amino acids of the corresponding protein (CFTR) were determined. The protein turned out to constitute a chloride channel embedded in the membrane of glandular epithelial cells. 70% of the mutant CF genes were found to lack three base pairs which code for the amino acid phenylalanine (Phe). Subsequent research revealed that the protein lacking this particular amino acid failed to be processed normally in the endoplasmic reticulum and as a result never reached and got built into the cell surface membrane. People carrying the mutated alleles in a homozygous state showed a complete lack of ion channels in their plasma membrane.” 1. Write the letters of the correct statements into the empty boxes. The glucose tolerance test is a standard method to test for diabetes. The blood samples of three individuals (A, B and C) were tested. They each consumed 1g of glucose per kilogram of body weight dissolved in water on an empty stomach. Then each had their blood sugar content measured at 30-minute intervals. The results are shown in mmol/litre in the graph. According to the data obtained person ‘C’ suffers from diabetes.
