    Course Prerequisites : BIY102 Fundamental BiochemistryObjectives:to prepare the student to gain knowledge on molecular mechanism of propagation and geneexpression.Expected Outcome:Students should be able to understand the principles of molecular biology and apply in the fieldof genetic engineering, medicine, and understanding of life processes.

    Unit No. 1 Genetic Material 9 hoursConcept and organization of genetic material. Physical and chemical structure of DNA, de-naturation and re-naturation kinetics of DNA. Genome organization in prokaryotes and in

    eukaryotes. DNA packaging in nucleosome, chromatin and chromosome. Identification of dnaas genetic material. Central dogma concept.

    Unit No. 2 DNA replication 9 hoursSemi-conservative replication. Enzymes in DNA replication-prokaryotic dna polymerases andeukaryotic DNA polymerases, fidelity and precessivity of polymerases. Replication inprokaryotes-origin of replication, fork formation, complex replication apparatus, replication ofleading and lagging strand. Okazaki fragments. Elongation, termination of replication.Eukaryotic DNA replication. Inhibitors of replication.

    Unit No. 3 Transcription 9 hours

    RNA structure, types of RNA, Gene Structure, RNA polymerases, transcription in prokaryotes-initiation and elongation, promoters, termination of transcription. Eukaryotic promoters.Distinction between pro and eukaryotic transcription. Posttranscriptional processing andmodifications of rna-mRNA, t-RNA and r-RNA, rverse transcription.

    Unit No. 4 Translation 9 hoursProtien structure-folding of polypeptide chain, alpha helix and beta secondary structures.Genetic code and its feature, wobble hypothesis, uniniversality of genetic code. Translationinitiation, elongation and termination in prokaryotes. Translation in eukaryotes. Post translationalmodifications. Antibiotics-inhibitors of protein synthesis.

    Unit No. 5 Regulation of gene expression 9 hoursPrinciples of regulation. Cis acting sites and trans acting molecules, transcriptional regulation, Transcription factor, DNA response element, feedback inhibition and allosteric control. the lacoperon, the trp operon, regulation of m-RNA stability, regulation of translation, eukaryoticregulation. Role played by HATs, HDACs, methylases and demethylases in regulation of geneexpression.
