1) La phase G0• Phase juste après la mitose (n chromosome)
• Etat quiescent ou de non division.
• Majoritaire dans les organismes adultes.
• Seules quelques cellules ne rentrent pas en phase G0.

1) La phase G0• Absence de cycline.
• E2F inhibé par la protéine Rb.
• Absence de facteurs mitogéniques.
• Aucune réplication d’ADN, ni de mitose.
• La durée du stade G0 est variable d’un tissu à l’autre.

2) Transition de la phase G0 à la phase G12.1 Facteur de croissance
• Premier acteur de la transition G0/G1.
• Provient du milieu extracellulaire.
• Se lie au récepteur dimérisé.

• Changement de conformation du récepteur dimérisé.

2.2 Activation de Ras• Fixation d’un adaptateur protéique lié a la
protéine Grb2.• Grb2 active une protéine Sos.

• La protéine Sos active la protéine Ras.

2.3 Phosphorylation• Protéine Ras entraine l’hydrolyse de GTP en
GDP et transmet le signal aux MAP kinases. • Les MAP kinases activent d’autres MAP
kinases.

2.4 Mise en place du complexe d’initiation
• Les facteurs de transcription se fixent sur l’ADN double brin au niveau d’une TATA box.

• Ce sont ces facteurs de transcription qui permettent à l’ARN polymérase de transcrire les ARNm.

• Les MAP kinases vont permettre d’activer des facteurs de la transcription à travers l’enveloppe nucléaire par phosphorylation.

2.5 Transcription
• Parmi les gènes transcrits se trouve Le gène Myc permettant de réaliser la protéine du même nom.

• La protéine myc est un facteur de transcription de L’ ADN, elle va activer l’expression d’un grand nombre de gènes dont celui de la cycline D.

• Cette cycline D se fixe à CDK4 créant ainsi le complexe cycline D/CDK4 actif qui servira dans la phase G1.
