Participated actively in the propagation of parasites in vivo, including inoculation, collection and purification as well as subsequent molecular biology techniques, including DNA/RNA extraction, PCR, RT-PCR, qPCR, electrophoresis of various sorts, PCR product purifications from liquids or gels and cloning of PCR products into bacterial plasmids for high fidelity sequencing. Variety of methods within the bio-informatics package Geneious® to analyse the resulting sequencing results.

1. Barcoding (NSERC, AHSI, Engage): Using a mitochondrial genetic target (cytochrome c-oxidase subunit I), various coccidian parasites (Eimeria, Isospora, Cystoisospora, Toxoplasma, etc) were sequence characterized following development of group-specific PCR primer sets. The resulting sequence data were then available for species delimitation (identification) as well as molecular phylogenetic studies. For many of the parasites, these were the first mitochondrial sequences to be obtained and published. (Ongoing project).

2. Related research projects that utilize DNA barcoding with apicomplexan parasites from various hosts include the following:

Wildlife Tissue and Fecal Coccidiosis (NSERC, CCWHC [Doug/Claire]);

Zoo Cases (NSERC, MTZ [Dale, Iga, Paulette]); Coccidia of Various Reptiles (Wildlife Samples) –

NSERC with Seville/McAllister. (All Ongoing Projects)

3. Molecular Characterization of Adeleid Parasites (NSERC) – Mitochondrial Genome Sequencing: Whole genome sequencing and molecular phylogenetic analyses of the mitochondria of Hepatozoon and related adeleid parasites. These are the first investigations into the mitochondrial genome sequence and structure of these common, but poorly understood, blood parasites and relatives (Ongoing project).

4. Improvement of commercial live coccidiosis vaccines (NSERC, FedDev): Product improvement, immunogenicity and safety testing of coccidiosis vaccine, live oocysts in chickens and turkeys. Purification of vaccine lines of coccidia, molecular characterization of same,

efficacy testing (immunogenicity and safety testing) in vivo in large battery trials. 2012-2013, completed successfully.

Viability Testing (MITACS): In support of live vaccination programs against coccidiosis in commercial poultry, we are evaluating molecular, in vitro methods to determine the viability of stored oocysts of poultry coccidia. The current requirement of in vivo viability testing is cumbersome, expensive and imprecise. (Completed successfully).
