a Pembentukan poliketida terjadi melalui reaksi Claisen antar unit asetil-CoA (C2). Dua unit asetil-CoA membentuk asetoasetil-CoA. Pengulangan reaksi selanjutnya menghasilkan poly-b-ketoester. Poly-b-ketoester dapat pula terbentuk melalui kondensasi antara asetil-CoA dengan malonil-CoA.
