KATA PENGANTAR
Puji syukur kami sampaikan kehadirat Allah SWT, karena atas segala limpahan
rahmat, taufik, dan hidayah-Nya kami dapat menyelesaikan makalah ini sesuai dengan yang
diharapkan. Dalam makalah ini kami membahas tentang “Etilen dan Asam Absisat”.
Makalah ini kami buat sebagai tugas terstruktur mata kuliah “Fisiologi Tanaman”.
Dalam makalah ini kami menjelaskan mengenai pengertian etilen dan asam absisat dan
fungsi bagi tanaman dan juga membahas tentang peranan kedua hormon tersebut bagi
tanaman.
Dalam proses pembuatan makalah ini, tentunya kami mendapatkan bimbingan agar
kami dapat menyelesaikan makalah ini sesuai dengan harapan dan dapat terselesaikan tepat
pada waktunya. Untuk itu, kami sampaikan terima kasih kepada pihak yang bersangkutan

BAB I
PENDAHULUAN
1.1 Latar Belakang
Setiap tanaman pasti akan mengalami fase pertumbuhan dan perkembangan, pada
fase-fase tersebut tanaman akan mengalami perubahan secara fisiologis. Perubahan
fisiologis tersebut dapat dipengaruhi oleh adanya beberapa hormon yang ada pada
tanaman tersebut. Proses tumbuh dan berkembangnya suatu tanaman dapat dipengaruhi
diantaranya oleh adanya hormon auksin, giberelin, sitokinin, dan asam absisat, dimana
pada masing-masing hormon memiliki fungsi dan sifat masing-masing sehingga
memberikan pengaruh perubahan yang berbeda-beda pula pada tanaman.
Dengan adanya hormon-hormon tersebut proses pertumbuhan dan perkembangan
tanaman dapat berjalan dengan mudah. Sejak tanaman mulai tumbuh hormon-hormon
pada tanaman tersebut telah mulai aktif bekerja, dan mereka bekerja sesuai dengan waktu
dan peranan masing-masing. Seperti pada saat pertumbuhan, saat tanaman masuk fase
generatif, saat tanaman mulai berbunga ddan berbuah, dan saat tanaman mulai mengalami
pemasakan buah. Maka hormon-hormon tersebut akan muncul dan bekerja sesuai dengan
waktu dan fungsi mereka masing-masing. Oleh sebab itu perlu dipelajari lebih lanjut lagi
mengenai keberadaan hormon tersebut pada tanaman beserta fungsi dan pengaruhnya bagi
tanaman.
1.2 Tujuan
Sebagai kewajiban untuk memenuhi tugas terstruktur yang diberikan.
1.3 Manfaat
Mahasiswa dapat mengerti dan memahami serta menerangkan mengenai hormon
etilen dan asam absisat.

BAB II
TINJAUAN PUSTAKA
2.1 Sejarah Penemuan Etilen
Pada tahun 1910, laporan tahunan yang disusun oleh HH Cousins dalam Jamaican
Agricultural Department menyebutkan bahwa buah jeruk sebaiknya tidak disimpan
bersamaan dengan pisang saat dikapalkan, sebab suatu aliran dari buah jeruk akan
meyebabkan buah pisang menjadi masak sebelum waktunya. Laporan tersebut menjadi
penjelasan pertama bahwa buah-buahan melepaskan gas yang memacu pemasakan dan
menjadi dasar penelitian yang berkelanjutan ditahun-tahun berikutnya.
Seorang ahli fisiologi berkebangsaan Rusia, Dimitry N Neljubow (1876-1926),
adalah orang pertama yang menyatakan bahwa etilen mempengaruhi pertumbuhan
tumbuhan. Pada tahun 1901 ia mencirikan etilen di dalam gas bercahaya dan
menunjukkan bahwa etilen menyebabkan tiga respon pada kecambah kacang kapri, yaitu;
terhambatnya pemanjangan batang; semakin menebalnya batang; dan munculnya
kebiasaan untuk tumbuh mendatar. Selanjutnya perluasan helai daun terhambat serta
pembukaan normal bengkokan epikotil terhambat. Dan baru pada tahun 1934, R Gene
membuktikan bahwa etilen disintesis oleh tumbuhan dan menyebabkan proses pemasakan
yang lebih cepat yang artinya gas etilen tersebut dapat memberikan efek fisiologis pada
tanaman (Salisbury & Ross, 1995).
2.2 Biosintesis Etilen
Etilen merupakan satu-satunya hormon dalam bentuk gas pada saat suhu biasa
(Sinha, 2004) yang memiliki struktur kimia CH2=CH2+H2PO4-+Cl- biasa juga dalam
bentuk H2C=CH2. Etilen tidak berpengaruh nyata dalam pertumbuhan vegetatif tumbuhan
walaupun memiliki efek yang signifikan dalam perkembangan akar dan batang (Hopkins,
1999).
Etilen dihasilkan dari sintesis primer sebagai respon dari stres tumbuhan dan
kemungkinan diproduksi disebagian besar dalam jaringan yang mengalami senescence
(penuaan) atau ripening (pematangan). Etilen bisanya digunakan untuk mempercepat

pematangan pada buah pisang dan buah lainnya yang masih hijau (belum matang) dalam
perdagangan yang disimpan dalam kapal. Perlu diketahui bahwa etilen sering dihasilkan
ketika auksin dalam konsentrasi tinggi dalam jaringan tumbuhan (Hopkins, 1999).
Pada tumbuhan tingkat tinggi prekusor utama dalam biosintesis etilen ialah asam
amino metionin. Dalam prosesnya terdapat 3 fase, yaitu;
Pertama, proses ini meliputi konversi metionin menjadi S-Adenosyl methionine
(SAM). Metionin diaktifkan oleh ATP dan enzim SAM sintase.
Kedua, SAM dipecah menjadi 1-amino cyclopropane-1-carboxylic acid (ACC)
dan 5’ metil tioadenosin. Prosesnya dikatalisis oleh enzim ACC sintase. Enzim ini
menjadikan pyrodoxal phosphate sebagai kofaktornya. ACC juga diubah yang kemudian
disimpan dari ethylene-N-malonyl ACC. Bentuk ACC mengontrol produksi etilen. Fase
ini langsung berkorelasi dalam pembentukan etilen. Faktor yang menghambat aktivitas
enzim ACC sintase juga menghambat biosintesis etilen. Auksin, factor stress (luka, water
stress, patogen), pematangan buah, gugur daun dan bunga mempercepat aktifitas enzim
ini, oleh karenya biosintesis etilen juga meningkat. Bahan kimia seperti aminoethoxy
vinyl glysine (AVG) dan amino o$xyacetic acid (AOA) menghambat biosintesis etilen
dari yang menghambat aktifitas enzim ACC sintase.
Ketiga, fase ini melibatkan oksidasi ACC yang menghasilklan CO2 dan HCN.
HCN dibentuk menjadi asam formic dan ammonia. Enzim yang berperan dalam fase ini
ialah enzim ACC oksidase yang mana melibatkan Fe++ dan askorbat untuk aktifitas ini.
Fase ini membutuhkan cahaya dan O2. Aktifitas enzim ini dipacu oleh pematangan,
pelukaan, dan dihambat oleh sulfhydryl inhibitors ( Co++, Cu++, Zn++). Oksidasi ACC tidak
rate limiting tidak seperti ACC sintase.
Regenerasi metionin muncul dalam sintesis siklus lanjutan dari 5’methyl
theoadenosine yang disebut “Siklus Yang” seperti terlihat pada gambar 1.
Lintasan lain dalam biosintesis etilen dalam tumbuhan adalah lintasan acrylic acid.
Disini, acrylic acid merupakan prekusor langsung dari etilen. Acrylic acid disintesis dari
asetat dan β-alanin (gambar 2).

Sintesis etilen meningkat dengan meningkatnya suhu yang mencapai 35o. dan
menurun ketika suhu mencapa 0-2oC. sintesis etilen dihambat oleh rendahnya konsentrasi
O2. Konsentrasi O2 yang tinggi tumbuhan tidak respon terhadap etilen (Sinha, 2004).

2.3 Katabolisme Etilen
Ethylen is metabolized to ethylene oxide in plants and micro-organism, wich is
incorporated into glyoxylate cycle as ethylene glycol and malonyl ACC. Ethylene may
also form a conjugate with glucose (Sinha, 2004).
2.4 Mekanisme Kerja Etilen
Many theories have been proposed to explain the mechanism of action of ethylene
of wich two are discussed below:
a. Membrane Permeability Theory: Ethylene acts by changing permeability of the cell
membrane. Ethylene is lipid soluble, hence, it gets dissolved in lipid of membrane and
alters the permeability of membrane. Increase RNA and protein synthesis. It
accelerates the secretion of α-amylase enzyme in barley aleurone layer but latter
synthesis is not enhanced. This result in hydrolysis of starch into sugars. High
permeability increase respiration, thereby, hastening fruit ripening process.
b. Molecular Level Action of Ethylene Theory: Ethylene acts at molecular level by
synthesizing a ccell wall softening enzyme polygalacturonases. Anthocyanidin is
formed by enzyme phenyl alanine ammonialyase which imparts bright colour to fruits.
Other enzymes also get increased (e.g. phenyl alanine ammonia, cellulose, polyphenol
oxidase, phospholipase, peroxidase). Cellulose perform softening of fruits during
ripening. Phospholipases causes degradation of phospholipids. Inhibitors of nucleic
acids/protein synthesi also inhibit ethylene action (Sinha, 2004).
2.5 Efek Fisiologi Etilen
(i) Germination and sprouting; Ethylene stimulates seed germination in barley
and other cereals. It stimulates the action of hydrolytic enzymes in storages tissues. This
causes sprouting and germination in dormant potato tubers, dormant stolons, corms and
rhizomes.
Ethylene breaks dormancy when it is exposed for some time, longer duration
might inhibit further growth of sprouts.

(ii) Formation and growth of roots; Both auxins and ethylene induce root
formation in mung bean hypocotyls cuttings. Ethylene treatment in some plants promotes
the formation of secondary roots and root hairs.
Ethylene stimulates lateral enlargement of roots (e.g. radish) but not the elongation
of root. Root elongation is inhibites by ethylene.
Geotropical bending of root occurs due to inhibition of root elongation caused by
ethylene. More ethylene is produced on the geotropically lower side of the root because of
more auxin contents. Ethylene intervenes the lateral transport of auxins, as a result auxin
is symmetrically distributed in plants. Hence, tropic movement fails and horizontal
growth occurs due to ethylene.
(iii) Shoot growth; Ethylene inhibits the longitudinal growth of stem but in turn it
causes lateral groth of stem which leads to increase in girth of stem. The lateral swelling
is due cell enlargement and not due to cell division. Ethylene mediated inhibition is
transitory, plants resume normal growth if ethylene is removed. Ethylene mediated
inhibion can also be overcome by CO2 application. Ethylene inhibits internode elongation.
High concentration of ethylene generally inhibits growth but it promotes growth in
paddy plants. Paddy plant produce more ethylene. The latter in turn promote growth in
former, which helps the plants in emerging out of submerged water.
(iv) Apical dominance; The growth of lateral buds is inhibited by ethylene just
like auxin, hence it causes apical dominance.
(v) Epinasty (Bending of leaves); Epinasty is process in which swelling of cells
on the upper part of the petiole occurs, which result into drooping of leaves. Monocots do
not show epinasty. Young leaves are more sensitive than old. Epnasty may be due to
migration of auxin to the upper side of the petiole. Higher auxin content result in
elongation of cells of the upper side of petiole. Hence, more growth occurs on upper side
which causes epinasty.
(vi) Flowering; Ethylene inhibits flowering in plants almost universally. However
pine-apple, plumbago, cucurbits are exeptions. Ethylene enhances pistillate flower

formation in cucurbits. Inn plumbago and pine-apple ethylene application in vegetative
body causes uniform flowering. Ethylene induces flowering S.D. plants even in long day
condition.
(vii) Sex expression; Ethylene application reduces the male flower formation and
favours female flower formation. As a result, more fruits are produced by ethylene
treatment but is size of fruits is smaller. Example – cucumber, pumpkin, ridge gourd,
melon etc.
(viii) Fruit ripening; Ethylene induces the typical fruit set. Is hastens the ripening
of harvested climacteric fruits like banana, mango, melon. Climacteric fruits are such
fruits which shoe increase in respiration after harvesting abruptly and then decline, while
non-climateric fruits (like grapes, lemon, orange) show decrease in respiration gradually
after harvesting. These fruits do not respond to ethylene.
During ripening ethylene increase cell permeability, which result into softening of
fruits as well as increases cell respiration.
(ix) Abscission; Ethylene accelerates the abscission of vegetative parts (leaves,
stems) as well as reproductive organs (flowers, fruits) of plants. The chief role of ethylene
is to promote senescence in leaves and induction of cell wall degrading enzymes in
abscission region. Ethylene further prevents auxin to reach the abscission zone, as a result
auxin level is reduced in abscission zone.
Older leaves produce more ethylene than young leaves. High concentration of
ethylene along with low concentration of auxin is responsible for abscission of old leaves.
Ethylene is absorbed by potassium permangate crystals. Thin chemical is kept with
flowers to keep the flowers fresh during transit (Sinha, 2004).
2.6 Hubungan Etilen dengan Auksin
Kemampuan IAA dan semua auksin tiruan untuk menaikkan produksi etilen
menimbulkan pertanyaan: benarkah banyak efek auksin betul-betul disebabkan oleh
etilen? Memang tampaknya etilen bertanggung jawab dalam beberapa kasus, termasuk
epinasti daun, penghambatan pemanjangan batang dan daun, induksi pembungaan pada

bromelia serta mangga, penghambatan pada pembukaan bengkokan epikotil atau hipokotil
kecambah dikotil, dan bertambahnya persentase bunga pada tumbuhan monoseus. Juga,
pelepasan auksin oleh serbuk sari yang berkecambah di stigma mendorong produksi
etilen; etilen ini menyebabkan penuaan bunga pada beberapa spesies. Gugur daun, bunga,
dan buah melibatkan interaksi antara auksin, etilen, sitokinin, dan asam absisat. Walaupun
demikian, pemacuan pertumbuhan, tahap awal pembentukan akar liar, dan banyak efek
auksin lainnya tampaknya tak bergantung pada produksi etilen. Hanya pada beberapa
bagian tumbuhan tertentu, dan hanya bila konsentrasi auksin cukup tinggi, maka produksi
etilen berperan cukup besar pada efek tertentu auksin (Salisbury & Ross, 1999).
2.7 Nilai Ekonomi Etilen
Ethylen has been used for synchronised flowering and fruit ripening for centuries.
Several chemicals which release ethylene are used now-a-days for promotion of
flowering in pine apple. Ethrel or Etepon (asam 2-kloroetilfosfonat) is ethylene releasing
chemical. It rapidly breaks down in water to produce ethylene.
Ethylene is also used for induction of feminity in cucurbits. Ethylene is widely used
in fruit ripening, improvement of colour (quality). Other ethylene releasing chemicals are
– BOH, ethypropyl phosphate, monoethyl sulphate. Ethylene is antagonist to auxin which
can be evident by following illustration: (i) Auxin delays abscission while ethylene
promotes. (ii) Auxin promotes elongation while ethylene inhibits (Sinha, 2004).
2.8 Sejarah Penemuan Asam Absisat (ABA)
Pada tahun 1963, asam absisat pertama kali dikenali dan dicirikan secara kimia di
California oleh Frederick T Addicott dan beberapa pembantunya, yang saat itu sedang
mempelajari senyawa yang menyebabkan gugurnya buah kapas. Mereka menamakan
salah satu senyawa aktifnya absisin I dan senyawa kedua (yang jauh lebih aktif) absisin
II. Absisin II ternyata ABA. Pada tahun yang sama, dua kelompok peneliti lain
menemukan ABA juga. Satu kelompok dipimpin oleh Philip F Wareing di Wales; mereka
mempelajari senyawa yang menyebabkan dormansi pada tumbuhan berkayu, khususnya
Acer pseudoplatamus. Mereka namakan senyawa yang paling aktif itu dormin. Kelompok
lainnya dipimpin oleh RFM van Stevenick, mula-mula di New Zealand, kemudian di

Inggris; mereka meneliti senyawa yang mempercepat gugurnya bunga dan buah pada
lupinus kuning (Lupinus luteus). Karena terbukti (pada tahun 1964) bahwa dormin dan
senyawa dari lupinus sama dengan absisin II, para ahli fisiologi bersepakat, pada tahun
1967, untuk menamakan senyawa itu asam absisat. ABA tampaknya umum ditemui pada
tumbuhan berpembuluh; juga terdapat di beberapa jenis lumut, ganggang hijau,
cendawan, namun tidak pada bakteri (Salisbury & Ross, 1999).
2.9 Biosintesis ABA
ABA is a terpenoid involved primarily in regulating seed germination, inducing
storage protein synthesis, and modulating water stress.
Unlike auxin, gibberelin, cytokinin hormone,
ABA as a single compound (Gambar 3). Originally
thought to be involved in regulating both abscission
and bud dormancy, ABA now appears to have little
to do with either of these phenomena. Two major
areas of ABA action appear to be in the mobilization
of reserves during seed development and germination
and in the response of leaves to water stress. ABA is known to induce transport of
photosynthate toward developing seeds and to promote the synthesis of storage protein.
During germination of cereals grains, ABA antagonizes the promotory effect of
gibberellins on α-amylase synthesis. Relatively large amounts of ABA are rapidly
synthesized in the leaves in response to water stress, where it appears to play a major role
in regulating stomatal opening and closure (Hopkins, 1999).
There are two pathway of abscisic acid synthesis:
Isoprenoid pathway: ABA is synthesized from mevalonic acid. Gibberellins
is also synthesized from mevalonic acid, i.e. both GA and ABA is
synthesized from common precursor.
Oxidation of carotenoids: Xanthophyll and vialaxanthin are precursors of
ABA in bean tissues. Oxidative cleavage of xanthophylls either by

photooxidation or by action of enzyme lipoxygenase produces xanthoxin and
aldehyde. This aldehyde is oxidized to produce ABA (Sinha, 2004).
Skema kedua lintasan tersebut dapat dilihat pada gambar berikut.

2.10 Katabolisme ABA
ABA forms ester with glucose. This ester is quite stable in plants and possesses
hormonal activity similar to ABA.
ABA + Glucose → ABA-glucose ester
ABA is metabolized to phaseic acid and dihydrophaseic acid in some plant. Seperti

2.11 Mekanisme Kerja ABA
Different theories have been proposed to explain the possible mechanism of action:
(i) Membrane Permeability Theory; Rapid action of ABA is stomatal closing and
inhibition of auxin mediated growth elongation in coleoptiles. These responses occur
due to change in membrane permeability of cells. The stomatal closing is observed
within a minute of ABA application. Likewise, inhibition of elongation of growth in
coleoptiles is observed in few minutes. ABA can also influence ion transport across the
membrane by inhibiting ATPase activity. Permeability of membrane might have been
changed due to decrease in unsaturated fatty acids (linoleic acid, linolenic acid) of the
membrane and increase in saturated fatty acids (palmitic acid). Increase in saturated fatty
acids favours increase in viscosity of membrane, thereby, reducing permeability of the
membrane, and consequently less diffusion of ions/molecules along the membrane.
(ii) Gene Expression Inhibition Theory; Long term effects of ABA take place at
molecular level. Such responses occur late after stimulus application (ethylene action).
Ethylene decrease transcription or translation process. Decrease in transcriptional rate
may be either due to decrease in RNA polymerase activity or increase in ribonuclease
activity. Inhibition of RNA synthesis can be detected after three hours of ABA

treatment. In bean seedling, two hours treatment with ABA decrease RNA synthesis by
80 per cent.
ABA may inhibit formation of certain enzymes without inhibiting total protein
synthesis. Ihle and Dure III (1972) showed that ABA inhibited production of protease
and isocitrase during germination of cotton seeds. This was due to inhibition of
translation of pre-exiting mRNA.
ABA inhibits the translation of amylase mRNA which inhibits the formation of α-
amylase in barley aleurone layers.
(iii) Secondary Messenger Involvement Theory: This theory assumes that ABA
acts by involvement of secondary messenger such as calcium and calmodulin. Ca2+ is
involved in ABA stimulated geotropism in maize roots. According to Coccuci and
Negrini (1989), ABA inhibits germination by controlling the development of some
activities dependent on calcium-calmodulin system. Stomatal closing by ABA also
requires Ca2+. In absence of Ca2+ ABA fails to close stomata (Sinha, 2004).
2.12 Efek Fisiologi ABA
Hormon ABA pada tumbuhan menimbulkan fisiologis bagi perkembangan dan
pertumbuhan tumbuhan, berikut diantaranya.
Inhibition of seed germination, seedling growth and bud growth: Exogenous ABA
inhibits germination of most non-dormant seeds. It might be due to inhibition of
enzymes protease and amylase along with carboxypeptidase synthesis. ABA also
inhibits water uptake in germinating seeds. Shortage of water in germinating seed
might inhibit their germination.
Seedling growth inhibition by ABA occurs due to decrease in water potential.
Exogenous ABA induces seed dormancy in woody plants. ABA content in dormant
bud is high. Hormone inhibits lateral growth of buds.
ABA acts opposite to growth promoting hormones. It counteracts the growth
stimulatory effects of auxins, gibberellins, and cytokinins. ABA is specific antagonist
of GA hence ABA is knoen as “Anti-gibberellin”.

Dormancy: ABA induces dormancy of seed and buds. After removal of ABA from
plant tissues, growth resumes. ABA inhibits the sprouting of potato buds completely,
thereby, maintaining quality of potato (sprouting makes potato sweet due to coversion
of starch into sugar).
Senescence and abscission: ABA accelerates senescence and abscission of leaves,
flowers, fruits. It promotes callus tissues development at abscission zone of petioles.
Stomatal closing: ABA is involved in stomatal closing. Increased content of ABA
favours stomatal closure, thereby, reducing transpiration rate. Hence it is regarded as a
good “antitranspirant”. ABA is increased during water stress. Response of ABA is fast
and occurs within a few minutes of ABA application. ABA inhibits K+ transport from
outside to guard cell and release malate from guard cells. This leads flaccidity in guard
cells. Consequently, stomata get closed.
Other roles of ABA: It inhibits flowering in long-day plants. ABA treatment inhibits

KesimpulanEtilen merupakan satu-satunya hormon dalam bentuk gas yang dihasilkan oleh
tumbuhan dan berperan dalam proses penuaan pada organ tumbuhan.
Asam absisat (ABA) merupakan hormon senyawa tunggal yang dihaslikan oleh
tumbuhan dan berperan sebagai hormon antagonis yang mempengaruhi fisiologis tumbuhan.
