CytoTune - iPS 2.0 Sendai Reprogramming Kit 는 sendai virus 를 이용하

여 Oct4, Sox2, Klf4, cMyc 유전자를 안정하고 편리하게 somatic cell에 전달하여

iPSC를 제작하는데 사용하실 수 있습니다. Sendai virus vector는 cytoplasm에서

replication이 일어나므로 핵막을 파괴하고 핵 안으로 들어가지 않으며 host genome

에 integration 되지 않아 다른 genetic information에 영향을 주지 않는 장점이 있습

니다. Cycotune 2.0 버전은 기존 Cytotune보다 reprogramming 효율이 2배이상

높으며, 렌티, 레트로바이러스에 비해서는 20~200배 이상 높은 효율을 나타냅니다.

Epi5 Episomal iPSC Reprogramming Kit는 야마나카 실험실의 Okita박사가 제작한 episomal vector로써Oct4,Sox2, Klf4,

L-myc,Lin28,p53DD 를 이용해 transgene-free,virus-free조건으로 iPSC reprogramming 효율이0.004~0.3%로 기존

episomal system 보다 높게 나타납니다.Epi5 Episomal iPSC Reprogramming Kit를 somatic cell 에 transduction 하기 위해

Differentiated neurons from human pluripotent stem cell (hPSC)–derived neural stem cells (NSCs) enable scientists to study human neural diseases from a large and diverse patient population like never before. However, during and after differentiation, the neuronal cell cultures are typically contaminated with proliferating neural progenitor cells that form clumps and make it nearly impossible to perform critical downstream assays on mature neurons.

Gibco™ CultureOne™ Supplement can be added to any conventional neuronal differentiation medium to eliminate more than 75% of contaminating neural progenitor cells with minimal cell death and no effect on kinase-mediated pathways. The resulting superior cultures of evenly distributed, differentiated neurons enable improved downstream assays, accelerated neuronal maturation, and maintenance in culture for 5 weeks or more.

hiPSC- or embryonic stem cell (ESC)–derived NSCs

Superior cultures consist of differentiated neurons, without progenitor cells and clumps.

Superior neuronal cell cultures can be easily maintained for 5 weeks or more.

Cultures consist of progenitor cells (SOX1) and differentiated neurons (MAP2) with clumps.

Contaminating neural progenitor cells form clumps that eventually peel off the culture plate.

Differentiated neurons from human pluripotent stem cell (hPSC)–derived neural stem cells (NSCs) enable scientists to study human neural diseases from a large and diverse patient population like never before. However, during and after differentiation, the neuronal cell cultures are typically contaminated with proliferating neural progenitor cells that form clumps and make it nearly impossible to perform critical downstream assays on mature neurons.

Gibco™ CultureOne™ Supplement can be added to any conventional neuronal differentiation medium to eliminate more than 75% of contaminating neural progenitor cells with minimal cell death and no effect on kinase-mediated pathways. The resulting superior cultures of evenly distributed, differentiated neurons enable improved downstream assays, accelerated neuronal maturation, and maintenance in culture for 5 weeks or more.

hiPSC- or embryonic stem cell (ESC)–derived NSCs

Experience superior neuronal cell culturesImproved downstream assaysCultureOne Supplement enables improved imaging, RNA expression, and electrophysiology assays. After 2 weeks of differentiation with CultureOne Supplement, images show evenly distributed, differentiated neurons (MAP2+) with greater than 75% reduction in NSCs (SOX1+) and cell clumps compared to conventional differentiation methods (A). These cells had increased neuronal mRNA expression, reduced NSC mRNA expression (B), and exhibited higher spike rates as measured by multi-electrode array (MEA).

Accelerated maturationNeurons differentiated from NSCs with CultureOne Supplement showed an increase in cytosolic calcium when depolarized with KCl (C), meaning they express significantly higher numbers of voltage-gated calcium ion channels, which are an important marker for neuronal

maturity and excitability. This, along with the longer neurites after 2 weeks of differentiation, demonstrates that CultureOne Supplement accelerates neuronal maturation.

Superior enrichment methodNeuronal cell cultures differentiated with CultureOne Supplement were compared to antimitotic enrichment methods including Ara-C and

mitomycin C (D). Cultures treated with CultureOne Supplement had improved neurite outgrowth, and reduced cell clumping and death. After 2 weeks, CultureOne Supplement yields an enriched culture of neurons at approximately 60% of the starting NSC plating density.
