Maligne Lymphome
Retrospektive Analyse zu R-Gemcitabin/Oxaliplatin beim rezidivierten DLBCL - 140 konsekutive Patienten am MSK New York von 2007 - 2018
(Schade et al, # 2904) ORR 26 %, mittleres PFS 1,4 Monate, mittleres OS 7,8 Monate -> insgesamt enttäuschende Daten
Background • Mosunetuzumab (RG7828; BTCT4465A)
– Full-length, fully humanized IgG1 bispecific antibody1
– Redirects T cells to engage and eliminate B cells; T-cell activation, cytokine elevation and increase in TILs observed (Hernandez et al. ASH 2019 P-1585)
– No ex-vivo T cell manipulation required (‘off-the-shelf’ and no delay in treatment)
• GO29781 – Phase I/Ib dose-escalation and expansion study in heavily pre-treated R/R B-cell NHL – Cycle 1 step-up dosing: mitigates CRS, allowing dose escalation to maximize therapeutic potential2,3
• We report data for 270 R/R B-cell NHL pts, including 30 pts with prior CAR-T
Registry number: NCT02500407 CRS, cytokine release syndrome; NHL, non-Hodgkin lymphoma; pts, patients; R/R, relapsed or refractory; TILs, tumor-infiltrating lymphocytes

Maligne Lymphome
• Mittleres Follow-up von 70,2 Monaten

-> Patienten mit EZH2-Mutation mit R- oder G-CHOP behandeln?

ZUMA-2 Disposition
• KTE-X19 was successfully manufactured for 71 patients (96%) and administered to 68 (92%) • Median time from leukapheresis to delivery of KTE-X19 to the study site was 16 days
PD, progressive disease.
34
Conditioning Chemotherapy (n = 69)
Enrolled/Leukapheresed (N = 74)
Received KTE-X19 (n = 68)
• Data cutoff: July 24, 2019 • Primary efficacy analysis (n = 60)
according to the protocol was conducted on the first 60 treated patients
• Safety analysis was conducted on all
treated patients (n = 68)
• Patients not treated (n = 5) ‒ Manufacturing failure (n = 3) ‒ Death due to PD (n = 2)
• Patients not treated after conditioning chemotherapy, found ineligible (n = 1)
Wang et al ASH 2019 Abstract 754
Representative PET Scans of Complete Response
• 50-year-old male patient with 3 prior therapies who presented with multi-compartmental MCL
• With KTE-X19, he achieved PR at Month 1 and CR at Month 3 and remains in remission 18 months later
CR, complete response; MCL, mantle cell lymphoma; PET, positron emission tomography; PR, partial response.
35
Wang et al ASH 2019 Abstract 754
Progression-Free Survival and Overall Survival
• Median PFS and median OS were not reached after a median follow-up of 12.3 months
OS, overall survival; PFS, progression-free survival.
36
