• Es wird gewartet, bis die Matrixwolke sich verdünnt hat und weniger dicht ist (ca. 10 ns)
Lösung: Methoden zur Energiekompensation
• Dann wird die Beschleunigungsspannung eingeschaltet, die Ionen werden beschleunigt.
• Ergebnis: • weniger Stösse der Ionen mit der Matrix, dadurch
weniger Energieverlust bzw. schmalere Verteilung der kinetischen Energie der Ionen
• Ionen, die aufgrund ihrer höheren Driftgeschwindigkeit weiter in IS1 eingedrungen sind, bekommen nicht mehr die volle Beschleunigungsspannung. Sie werden am Detektor von den Ionen eingeholt, die eine geringere Anfangsdrift hatten.

Lösung: Methoden zur Energiekompensation

MALDI-TOF Linear vs. reflector mode

Wenn der Reflektor-Mode im TOF so viel bessere Massenauflösung liefert: warum verwendet man überhaupt noch lineare Systeme? Der lineare Modus hat Vorteile • bei labilen Molekülen: durch den MALDI Prozess werden sie angeregt
und zerfallen u.U. In der Driftstrecke. Sie werden dann nicht mehr richtig reflektiert und gehen verloren (Beispiele: Zucker mit Sialinsäure)
• bei großen Moleküle mit MW>30 kDa: sie zeigen oft eine breite Energieverteilung, die der Reflektor nicht mehr kompensieren kann. Hier leidet die Transmission, d.h. grosse Teile des Ionenpaket gehen verloren, die Empfindlichkeit wird gering.
Es ist sinnvoll, beide Modi im TOF einzusetzen, linear und Reflektor, zwischen denen man je nach Bedarf schalten kann.

Problem: Wenn die Ionen nach Verlassen der Beschleunigungsstrecke zerfallen oder durch Stöße fragmentiert werden, verlieren sie einen der reduzierten Masse entsprechenden Teil ihrer kinetischen Energie
z.B. ist bei der intakten Masse 1000 Da (Beschleunigung mit 10 kV): Intaktes Ion bei 1000Da Ekin intakt= 10 keV (100%) Fragment f1 mit 500Da Ekin f1 = 5 keV (nur noch 50%) Fragment f2 mit 100Da Ekin f2 = 1 kV (nur noch 10%)
