分子量(MW)が異なるペプチド・タンパク質の分離を0.1%TFAを含む溶離液条件下にて、40℃と70℃で比較しています。
分子量が大きいペプチド・タンパク質の分離には溶離液へのイソプロパノール(IPA)の添加がピーク形状の改善に有効ですが、分子量が10,000以上の
場合、40℃においてはブロードなピークとなっています(condition B 上図)。
40℃と比較して70℃の高温条件では分離選択性の変化やピーク形状の改善により分離度が向上し、この効果は特に分子量が大きい場合に顕著です。
一般的に大きい分子は小さい分子よりも拡散速度が遅いため、ピークがブロードになる傾向があります。高温条件では溶離液の粘度の低下と物質移動
速度の改善によりピーク形状が改善されます。
ペプチド・タンパク質の分離において、カラム温度の変更は分離度の向上に有効です。
ヘモグロビンのトリプシン消化物の分離においてカ
ラム温度を40℃から70℃に上げることで、ピーク
キャパシティ(分離可能ピーク数)が23%向上して
います。
粒子径1.9 μm、長さ100 mmのTriart C18カラムを
2本連結することで分離は劇的に向上しています。
また、高温条件のため、総カラム長200 mmながら
多くのUHPLC用システムの耐圧より十分に低圧
力で分析可能です。このような手法は、ペプチド
マッピングなど高分解能が要求される分離に有効
です。

HPLC共通条件
抗菌ペプチドの分離最適化例
一般的なペプチド分析条件で検討する
と分離しませんが、温度を70℃に上げ
て分析すると1, 3のピークと2のピーク
が分離しています。
TFAを含む溶離液を使用した高温条件はペプチド・タ
ンパク質の分離の向上に有効ですが、通常の逆相カ
ラムではカラム寿命が問題となります。
Triartは新開発の有機シリカハイブリッド基材に緻密
な表面修飾を施しているため、左図の1%TFA、70℃
のような厳しい条件でも卓越した耐久性を有していま
す。このため、通常のカラムには適用しにくい条件で
の分析も可能です。
有機溶媒の組成をacetonitrileから2-
propanol/acetonitrile混液に変更し、グラジエ
ント条件を最適化することで、同等の分析時間
で分離度が向上しています。
ペプチド・タンパク質の分析では、移動相に溶
出力の高い2-propanolを添加することで分離
が改善することがあります。
