microRNA란?

microRNA는 non-coding RNA로 19 ~ 22nt 길이의 작은 RNA 단

편으로 유전자 발현 조절에 중요한 역할을 한다. Post-transcrip-

tional 단계에서 발현 조절에 관여 하는 것으로 알려져 있으며, 포유

류 단백질 발현 유전자의 60% 정도의 발현에 관여하는 것으로 생

각된다. microRNA는 암, 심장질환, 신경질환 등의 다양한 질병과

도 밀접한 연관을 갖고 있을 것으로 예상되어 많은 연구자들이 관

심을 갖는 분야 중 하나이다.

Exiqon의 독자적인 기술 LNA(Lock Nucleic Acid)란?

Locked Nucleic Acid(LNA)란 DNA/RNA 구조내의 2’ 탄소원자 위의 산소 원자와 4’ 탄소 원자를 고정시킨

(노란색) RNA analogs로 LNA를 포함한 oligonucleotide는 DNA나 RNA에 높은 결합력과 특이성을 나타낸

다. 이러한 특성을 지닌 LNA 기술을 적용할 경우 균일한 Tm값을 가진 probe 나 primer를 제작할 수 있다.

LNA의 장점

• 1 염기 수준의 mismatch 검출가능

• microRNA에 존재하는 2종의 family 간 식별이 가능

• 체액이나 FFPE 샘플 등에서 높은 검출 민감도로 해석 가능

• ng의 total RNA로부터 프로파일링 가능

• 단일 microRNA의 경우 1 pg의 total RNA로부터도 해석 가능

• GC 또는 AT 함유율에 관계없이 높은 재현성의 microRNA 검출 가능
