簡便・迅速な形質転換技術

・プラスミドDNAを保持する大腸菌や枯草菌を溶菌させ、溶菌液を用いて形質転換を行う方法を提供する。
概要
・ドナー宿主(大腸菌、枯草菌)を、ファージ感染や界面活性剤などを利用して溶菌させる。
・溶菌液を直接、または添加剤を添加後、レシピエント宿主(大腸菌、枯草菌、酵母など)と接触させる事によりプラスミドDNAをレシピエント細胞内に導入し形質転換を行なう。
・生化学的精製操作を経ず異宿主間のベクターDNA移動ができるため、迅速かつ簡便な遺伝子導入が可能。特に精製過程でダメージを受けやすいサイズの大きなDNAに対して効果的。
・多種類のDNAの評価・スクリーニングや長鎖DNA(ゲノム)合成をハイスループットに行うには極めて有効な方法。

Transport across the cytoplasmic mem-brane and DNA processing. In both B.subtilis and N. gonorrhoeae, similar poly-topic membrane proteins (ComEC and ComA,respectively) are required for DNA trans-port into the cytosol (8, 33). These large pro-teins (ComEC contains 776 residues) areproposed to form channels for the passage ofDNA (34). In addition, the Gram-positivesystems encode a membrane-bound ATPase,ComFA, that functions in DNA uptake (35).ComFA resembles the family of Asp-Glu-Ala-Asp (DEAD) box helicases, and mayassist the translocation of DNA through themembrane or carry out strand separation. InS. pneumoniae, the membrane-associatedEndA nuclease degrades the nontransform-ing strand, even when the ComEC equivalentis absent (10). In B. subtilis, the identity ofthe corresponding nuclease is unknown, butdegradation of the nontransforming strandseems dependent on passage through orinteraction with ComEC (12).
Cellular location of DNA uptake and therole of cytosolic proteins. In B. subtilis,which is a rod-shaped bacterium, DNAbinding and uptake take place preferentiallyat the cell poles, where the membrane-associated proteins ComGA and ComFAand the cytosolic ssDNA binding protein
YwpH colocalize (36). Several additionalcytosolic proteins participate in transfor-mation, and some have been shown to as-sociate with ssDNA entering the cell. In S.pneumoniae, the Smf protein protects trans-forming DNA from degradation in thecytosol (37). The repair/recombination pro-teins RecN and RecA also localize to thepoles of competent B. subtilis (38). RecNoscillates from pole to pole, but becomesstatic at one pole when transforming DNAis added. RecA localization depends onComGA; when DNA is added, RecA formsa filament extending from the pole to thecentrally located nuclear body, perhaps fa-cilitating the search for a homologous siteon the chromosome.
A transformation model. We propose thatrepeated cycles of Y-pilus assembly anddisassembly drive a DNA molecule throughthe cytoplasmic membrane channel formedby ComEC and that an unidentified DNAbinding protein anchors DNA to the Y-pilus.ComEA may ensure processivity by main-taining contact with DNA as these cyclespush DNA through the channel (8). Theproton motive force might drive disassemblyof the Y-pilus. Finally, the binding energy ofcytosolic ssDNA binding proteins mightprovide a pulling force by a Brownian
ratchet mechanism (39), and the helicase/translocase ortholog ComFA may also assistuptake.
ConjugationMost bacterial and some archaeal speciesencode conjugation systems, and severalclasses of mobile elements exist, includingself-transmissible and mobilizable plasmids,conjugative transposons, and integrative con-jugative elements (40). We will restrict thediscussion to a few of the better-characterized,plasmid-encoded conjugation systems ofGram-negative bacteria and draw on examplesfrom the Gram-positive bacteria where infor-mation is available.
The conjugation apparatus is composedof a cell-envelope–spanning translocationchannel and either a pilus for Gram-negativebacteria or surface-localized protein adhe-sins for Gram-positive bacteria (Fig. 1B)(41, 42). The mating pair formation (Mpf)proteins elaborate the extracellular pilus,and these subunits plus the coupling protein,here termed the substrate receptor, mediatesubstrate transfer across the cell envelope(43, 44). Agrobacterium tumefaciens elabo-rates a model conjugation machine from 11Mpf subunits, VirB1 to VirB11, and theVirD4 substrate receptor, to deliver onco-genic transferred DNA (T-DNA) to suscep-tible plant species (45). Many plasmidconjugation systems, exemplified by trans-fer systems of plasmids R388, F, and RP4,are built from nearly complete sets of VirB/D4-like subunits, whereas other systemshave only one or two discernible homologs(41, 42, 44–47). Conjugation and ancestrally-related translocation machines make up thelarge and functionally versatile family oftype-IV secretion systems (46–48).
Processing of the conjugative-transferintermediate. The processing of substrateDNA for conjugative transfer is a widelyconserved reaction among Gram-negativebacteria and unicellular Gram-positive bacte-ria (Fig. 1B) (49). A relaxase plus one ormore auxiliary factors initiate processing bybinding the origin-of-transfer (oriT) se-quence and cleaving the DNA strand des-tined for transfer (T-strand). The relaxaseremains covalently bound to the 5¶ end of theT-strand, resulting in the formation of therelaxase–T-strand transfer intermediate. Thisprocessing reaction clearly is distinct fromthe strand-specific degradation pathwaysoperating during transformation. Also incontrast to the competence systems, signalsconferring substrate recognition are carriednot by the DNA but by the relaxase; theseminimally consist of positively charged orhydrophobic clusters of C-terminal residuesand are found in other protein substrates aswell (50–52). ・その他のDNA導入法との比較
プラスミドDNAの抽出・精製操作を含まないDNA導入法として接合伝達法が挙げられる。しかし接合伝達法は一般的にドナー細胞とレシピエント細胞の相性や培養条件が限定されている場合が多く、現時点ではまだ汎用的な操作法としては向いていない。
3) 従来法(一般的なプラスミド精製→形質転換)との比較
従来法では、形質転換に用いるDNAは生化学的な抽出・精製が必要とされてきた。抽出・精製操作において、特に50kbを越える長鎖DNAは長大なポリマーであるため物理的ダメージを受けやすく取扱いが困難であった。

1)物質生産に必要な遺伝子群を含む長鎖遺伝子クラスターを酵母細胞に導入することで、抗体など高度な有用タンパク質の生産分泌可能な有用形質転換酵母を作製することができる。溶菌法は操作が簡便であるため、ハイスループットな運用が可能である。従って、ゲノムデザインサイクル(GDC)を活用し、物質生産に特化した形質転換酵母を作成する際は、本発明の溶菌法は重要な技術基盤ツールになると期待される。
2)大腸菌などで作成されたコンティグクローンによるDNAライブラリーを目的の宿主のゲノムに順次導入することでゲノム領域の再構築を実現する。溶菌法を用いることで長鎖DNA合成を簡便、迅速に操作することができる。

• 溶菌法は操作が非常に単純なため複数のサンプルを同時に扱うことに適している。現在、マルチウェルプレートを用いた運用が可能なところまで確認済み。

• オートメーション化については既存の機器を組み合わせることで開発できると見込まれる。培養細胞への適用については添加剤などの条件検討で克服できると見込まれる。
• また、有用宿主作成のための形質転換技術を開発中の企業、長鎖DNA(遺伝子クラスター)またはゲノム合成分野への展開を考えている企業には、本技術の導入が有効と思われる。
• 微生物や培養細胞を用いた有用物質生産の技術を持つ企業との共同研究を希望。
