細胞をベースとするアッセイ生存細胞とその環境は多岐にわたるため、すべての細胞種に適用できるアポトーシスや生存率、またはその他数値化アッセイを単一化するのは、不可能です。アポトーシスと生存率は単一の生理学的または形態学的パラメーターで定義するのは容易ではないため、酵素活性、細胞膜透過性、酸化還元（レドックス）能などのいくつかの指標を総合することが望まれます。それぞれのアッセイには長所と限界があり、実験に偏向がもたらされることがあるので、異なる用途には異なるアプローチがしばしば求められます。これらの理由のため、Invitrogenではアポトーシス、細胞周期の分析、細胞の生存能および増殖のアッセイに広範な選択肢を提供しています。これらのアッセイの多くは蛍光に基づいており、一般に放射性同位元素テクニックより危険性が低く安価で、比色法より感度が高く、動物試験法より簡便です。さらに、本章で述べるアッセイの多くは細胞別に分析でき、そのいくつかは顕微鏡、フローサイトメトリーまたはマイクロプレートリーダーによる検出にも同様に適しており、多くの場合ハイスループット用途に適しています。

アポトーシス細胞死に向かう規則正しい進行は多細胞生物の生命にとって欠くことができません。アポトーシス（遺伝的に制御された細胞の除去）は不必要な細胞や組織を有機体から除去することによって適切な成長と発達を可能にするだけでなく、免疫系やウイルスに感染した、またはDNAが損傷された余分な細胞を破壊することによって有機体に対する脅威を抑制します。プログラムされた細胞死は形態学的にも生化学的にも傷害による細胞死（壊死）とは異なります。壊死細胞とは対照的に、アポトーシス細胞はDNAの断片化やいくつかの細胞タンパク質の分裂または分解だけでなく、核クロマチンの圧縮、細胞質の収縮および細胞膜結合アポトーシス体の生成を示します。アポトーシスの空間的時間的成分を明らかにするため、これらの差異は蛍光プローブで選択的に標的にすることができます。癌や脳卒中、アルツハイマー病およびいくつかの自己免疫疾患では誤って調節されたアポトーシスが関係しており、これらの経路を解明する上でこれらのプローブは重要なツールになっています。細胞の生存能においても、単一のパラメーターですべてのシステムの細胞死を完全に定義することはできません。したがって、アポトーシス事象とその時間的関係を研究する際は多パラメータアプローチがしばしば有利になります。これらのプローブにより、特定の実験系におけるアポトーシスの相対的ステージを判定できるだけでなく、生存細胞とアポトーシス細胞および壊死細胞を区別できるようになります。単一のアッセイではアポトーシスを適切に分析できないことに留意してください。

クロロフィルおよびフィコビリソームからの天然の赤色自家蛍光を補うため、さまざまな青色または緑色蛍光プローブで染色したシアノバクテリアの画像。丸い細胞はSynechocystis sp.（PCC 6803株）で、細胞膜は緑色蛍光BODIPY®

アポトーシス誘導の時間経過の一例。時間調節誘導を用いて複数のマーカーを発現する細胞集団間の関係を比較することにより、あるモデルにおいてアポトーシスプロセスのさまざまな成分が明らかになる相対的順序を推定することが可能になる。当社では0-4時間にわたり10µM camptothecinで誘導したJurkat細胞を用いていくつかの試薬を検討している。上のチャートはannexin V

ホスファチジルセリンのトランスロケーション

アポトーシスを起こしている細胞では、ホスファチジルセリン（PS）が細胞膜の細胞質側から外層側に移動します。アネクシンVはPSに高い親和性を示し、通常、アポトーシスアッセイに使用されます。本節のVybrant®アポトーシスアッセイキットは全て、アポトーシス細胞の検出にアネクシンVの蛍光コンジュゲートを使用しており、死細胞とアポトーシス細胞の区別には別の染色剤を使用しています。蛍光を発し、ビオチン化したアネクシンVのコンジュゲートも単品として別々にご利用いただけます。

JurkatヒトT細胞白血病細胞は1μMのカンプトテシンで処理された。externalホスファチジルセリン（早期のアポトーシス細胞の特徴）は、Alexa Fluor® 488 annexin

V (A13201)で検出された。後期アポトーシス、壊死細胞は propidium iodide で染色された。

　2010年新製品のViolet Ratiometric Membrane Asymmetry Probe/Dead Cell Apoptosis Kitには比率測定膜対象性プローブのF2N12Sが入っています。F2N12Sは、PSの移動に伴う原形質膜の表面電荷の変化に応じて、2色の蛍光の比率が変わります（Shynkar, V.V., et al. J. Am. Chem. Soc. 2007. 129: 2187-2193）。アネキシンVと違って、このアッセイは特別なバッファーや洗浄ステップが不要です。またこのアッセイは、接着細胞の場合に、細胞をはがすことによる細胞膜の損傷の影響をより受けにくいため、より高品質のデータが期待されます。

モノマー・シアニン系染色

アポトーシスは、原形質膜にホスファチジルセリンを検出できる蛍光アネクシンVコンジュゲードによってアッセイをします。しかし状況によっては、その方法が最適でないことがあります。このアッセイは、アネクシンV結合に必要である高カルシウム濃度、サンプルの洗浄液、ホスファチジルセリン検出時のトリプシン処理による接着細胞等の悪影響をもたらします。モノマー・シアニン系染色(PO-PRO.-1, YO-PROR-1, TO-PROR-3)は、原形質膜の非対称性な損失を伴う透過性の変化によって浸透性アポトーシス細胞を検出できます。これらの色素は生細胞の膜は通常通過しません。これらの色素はアポトーシス細胞に入って核酸に結合します。この3種の色素は特有の励起波長を持ち、多重染色アッセイに適しています。

ミトコンドリアの機能

膜電位 ─ 固定可能なプローブ

MitoTracker®色素（特許保有）は、生細胞のミトコンドリアを染色する膜電位依存性プローブです。MitoTracker®色素の蛍光シグナルは膜が脱分極したミトコンドリアよりも活性を有するミトコンドリアの方が明るく、細胞群中の健常な細胞を識別する方法となります。MitoTracker®色素の染色パターンは、その後の免疫細胞化学、DNA末端標識、in situハイブリダイゼーション、対比染色全体を通じて保持されます。同じ実験に緑色蛍光色素とペアで使用することができ、スペクトルのオーバーラップが最小になるようなMitoTracker® Red CMXRos（最大励起波長579 nm、最大蛍光波長599 nm）を販売しています。近赤外色素を併用してお使いになる場合には、MitoTracker® Orange CM™Ros

をお選びください。MitoTracker® Red CM-H2XRosとMitoTracker® Orange CM-H2™Rosはバックグラウンドが低い実験に最適です。MitoTracker®色素の還元タイプは、細胞の細胞質に到達するまでは蛍光を発しません。これら色素は通常行なわれているパラホルムアルデヒド固定中には十分に保持されますが、染色パターンが変化する可能性があります。実験には適切な対照を使用しなければなりません。MitoTracker® Orange 色素とMitoTracker® Red色素由来の蛍光シグナルは、緑色蛍光タンパク質（GFP）と組み合わせて使うことができるので、GFPトランスフェクション細胞の染色に有用です。

膜電位 ─ 固定化しないプローブ

膜透過性色素は、アポトーシスの間に生じるミトコンドリアの膜電位の消失を迅速、簡単かつ確実に検出することができます。しかし、MitoTracker®色素とは異なり、これらの色素の染色パターンは固定中に失われてしまいます。これらの最初の色素であるJC-1は、陽イオン性色素であり、緑色（～ 525 nm）から赤色（～ 590 nm）への蛍光発色シフトによって示されるように、ミトコンドリア中に電位に依存して蓄積されるので、レシオメトリック測定（比率測定）に有用です。当社はまた、細胞毒性効果を誘発することなく、活性を有するミトコンドリアによって容易に隔離されるローダミン123とジヒドロローダミン123も提供しています7。このようなローダミン色素の取り込みと平衡化は、30分以上かかる他の膜電位感受性色素と比べると迅速です（数分間）8。緑色蛍光色素を取り込む多色用途（最大蛍光～ 520

nm）には、テトラメチルローダミンのメチルエステルあるいはエチルエステル（TMRMとTMRE）をお選びください。

ミトコンドリア遷移孔ミトコンドリアの外膜と内膜の成分によって形成される非特異的チャネルの透過性遷移孔は、アポトーシスと壊死性細胞死の間におけるミトコンドリア成分の放出に関与していると思われます。Image-iT® LIVEとMitoProbe™遷移孔アッセイキット（それぞれ蛍光顕微鏡用とフローサイトメトリー用）はミトコンドリア膜電位のみに依るアッセイと比較して、ミトコンドリア透過性遷移孔の開口を測定するためのより直接的な方法を提供します。これらのアッセイでは無色で蛍光を生じないエステラーゼ基質のcalceinのアセトオキシメチル（AM）エステルと、calcein蛍光の消光剤のCoCl2を用いてミトコンドリアを選択的に標識します。細胞にcalcein AMをロードすると受動的に細胞内に拡散し、ミトコンドリアなどのサイトソル部分に蓄積します。細胞内部ではcalcein AMは細胞内エステラーゼによって分解されて非常に極性の高い蛍光色素のcalceinを遊離しますが、これは比較的短期間にわたってミトコンドリアや細胞の膜を通過しません。CoCl2を添加するとサイトソルのcalceinの蛍光は消光しますが、ミトコンドリアのcalcein

からの蛍光は維持します。Image-iT® LIVEミトコンドリア遷移孔アッセイキットにはミトコンドリア画像の対比染色用としてMitoTracker® Red CMXRosも入っています。

チトクロームC放出の検出　アポトーシスの引き金となるもののひとつに、ミトコンドリアからのチトクロームCの放出があります。当社のSelectFX® Alexa Fluor® 488チトクロームCアポトーシス検出キットは、チトクロームCの検出に抗チトクロームC抗体とAlexa Fluor® 488標識二次抗体を使用しており、さらに、細胞の固定と透過性獲得に必要な保存液と、染色用プロトコールが含まれています。

MitoProbe™ JC-1アッセイキット（M34152）を用いたJurkat細胞のフローサイトメトリー分析。Jurkat細胞を5％二酸化炭素中37℃で15分間、2µMのJC-1で染色してリン酸緩衝生理食塩水（PBS）で洗浄し、488nmの励起光で530nmと585nmの帯域放出フィルターを用いてフローサイトメーターで分析した。未処置培養細胞は図Aに示した。図Bは37℃で4時間にわたり10µMの

Caspase活性化

生存細胞におけるCaspaseアッセイVybrant® FAM Caspaseアッセイキットは活性caspaseを検出するために、caspaseの蛍光阻害薬（FLICA™）に基づく新しいアプローチを採用しており、これは基本的に親和性標識です。FLICA™試薬はcaspase特異的アミノ酸配列を介してcaspaseのシステインと共有結合するフルオロメチルケトン（FMK）部分と結合を有します。このFLICA™試薬には蛍光レポーターとしてカルボキシフルオレセイン基（FAM）が結合されています。FLICA™試薬は認識配列を介して活性化caspaseの酵素反応中心と相互作用すると考えられ、FMK部分を介して共有結合します。FLICA™阻害薬は細胞透過性で細胞毒性がありません。未結合FLICA™分子は細胞外部に拡散するので洗浄して取り除きます。残った緑色蛍光シグナルが、阻害薬を添加した時点に存在していた活性caspase量の直接の指標です。

クロマチン凝縮

　アポトーシスが起こっている細胞では、核のクロマチン凝縮が増大することが示されます。幾つかの核染色剤で標識すると、このコンパクトな核は過度に蛍光を発します。Hoechst 33342は全ての細胞に容易に取り込まれるUV励起可能な核酸染色色素です。これは青色蛍光であり、アポトーシス細胞の凝縮された核では特に明るい蛍光を発します。一方、プロピジウムアイオダイドは原形質膜が損傷している細胞にのみ取り込まれます。このふたつの核酸染色剤を同時に使用した時の染色パターンから、健康な細胞群、アポトーシス細胞群、死細胞群をフローサイトメトリーまたは蛍光顕微鏡法で区別することができます。当社はこのふたつの色素を、単品もしくはキット（カタログ番号V13244）として販売しています。

細胞周期分析細胞内DNA含量および細胞周期の分布に関する生存細胞の研究は、腫瘍の挙動とサプレッサー遺伝子メカニズムの研究、アポトーシスのモニタリング、および物理的、化学的または生物学的状況の変動によってもたらされる増殖パターンの変動を検出する上で有用です。細胞は集団内で細胞周期の主に3つの相に分布しています：G0/G1期（細胞当たり染色体が1対）、S期（DNA合成期、DNA量は変動）およびG2/M期（細胞分裂前、細胞当たり染色体が1対）。DNA含量はDNA量と比例するシグナルを放出するDNA-選択的蛍光染色を用いて測定できます。次にこれらの染色された集団のフローサイトメトリー分析を用いて頻度ヒストグラムを生成すると、細胞周期のさまざまなステージが現れます。この分析は通常、透過または固定された細胞を対象に細胞不透過性の核酸染料を用いて行なわれますが、生存細胞を対象に細胞透過性の核酸染料を用いて行なうこともできます。

生存細胞の細胞周期分析


Vybrant® DyeCycle™は、生存細胞におけるDNA含量をフローサイトメトリーを用いて分析するために開発されました。Vybrant® DyeCycle™はすべてDNA選択的、細胞膜透過性で、二本鎖DNAに結合するまで蛍光を生じません。DNAに結合すると、これらの色素はDNA量に比例した蛍光シグナルを放出します。Vybrant® DyeCycle™による細胞の染色はシンプルで、懸濁させた細胞をVybrant® DyeCycle™の存在下でインキュベートして蛍光を直接測定するので、追加の処理や遠心分離は必要ありません。蛍光データを用いて頻度ヒストグラムを生成するとさまざまなステージの細胞周期が現れます。Vybrant® DyeCycle™は広く用いられている488nmと405nm励起光源の長所を利用しており、細胞周期の研究はすべてのサイトメトリーで行なえ、他のパラメーターについても細胞集団の同時染色が可能です。Vybrant® DyeCycle™ Violet

は405nmのレーザー光線に最も適合していますが、UV励起光源を用いることもできます。Vybrant® DyeCycle™ Green

はアルゴン-イオンレーザーの488nmスペクトル線で効率的に励起できます。Vybrant® DyeCycle™ Orangeは488nmまたは532nmのレーザー光線のいずれを用いても励起させることができます。またVybrant® DyeCycle™は細胞に対して無毒なので、細胞周期の相に基づいて細胞集団を分類することもできます。固定細胞で細胞周期を解析するのには、2009年新製品のFxCycleが使用できます。FxCycleはブルーレーザー励起色

素と併用できます。蛍光スペクトル幅が狭いので、488nm励起色素とのコンペンセーションがより少なくて済みます。

Click-iT™ EdU細胞増殖アッセイキットBrdUアッセイに代わる、迅速でより簡便な方法が得られます。EdU（5-エチニル-2'-デオキシウリジン）はチミジンのヌクレオシドアナログで、DNA合成中にDNAに取り込まれます。EdUの短時間パルスで反応させるだけで、DNAを活発に複製している細胞を標識することができます。より長いパルスでEdUを反応させると、活発に増殖している集団中のすべての細胞が標識されます。検出はクリック反応に基づいて行ないます。これは銅が触媒するアジドとアルキン間の共有結合です。この用途ではEdUにはアルキンが含まれ、Alexa Fluor® 488色素にはアジドが含まれます。次に標準的なフローサイトメトリー法を用いてS相にある集団中の細胞の割合を測定します。Click-iT™ EdUアッセイキット中のアジドはサイズの小さな色素なので、緩やかな条件を用いて細胞中のDNAを効率的に標識することができます。BrdU法とは異なりDNAを変性させる必要がないので、検出試薬は組み込まれたEdUと相互作用することができます。Click-iT™検出試薬がDNAに接近できるようにするためには、標準的なアルデヒドをベースとする固定と界面活性剤で透過性を増加させるだけで十分です。このように、EdU細胞増殖アッセイは取扱いが容易であるだけでなく、細胞周期用色素と両立でき、抗体をベースとする表面、細胞内マーカー検出を用いて、多重検出を行なうことができます。

アミン反応性色素による細胞標識、細胞増殖の世代解析蛍光色素のサクシニミジルエステル(SE)は現在最も広範囲に使用され

ている細胞の世代分析用プローブです。蛍光を発するSEは、リジン側鎖や他の反応可能なアミン基と反応して、自発的、非可逆的に細胞内タンパク質と細胞表面タンパク質両方に結合します。細胞が分裂する時、SE

標識は娘細胞に等しく分配されるので、親の蛍光の半分になります。その結果、増殖細胞群の連続する世代は、細胞蛍光強度が半分になることによってマークすることができます。これはフローサイトメトリーによって容易に追跡することができます。この用途に最も一般的に使用されている色素は、カルボキシフルオ

レセインジアセテート、サクシニミジルエステル(5(6)-CFDA SEまたはCFSE)です。CFDA SEでは細胞標識はより均質です。その結果、膜マーカーPKH26のような他の細胞追跡色素に比べ良好に世代間を分けることができます。CFDA SE標識のフローサイトメトリー分析を用いて、リンパ球の8～ 10連続世代を解析することができます。移植された細胞では、何週間もin vivoでCFDA SEのシグナルを追跡することができます。

細胞生存率と活動性生存細胞とその環境は多岐にわたっているため、すべての細胞種に適用できる単一の生存率アッセイを工夫するのは不可能です。生存率は単一の生理学的または形態学的パラメーターで定義するのは容易ではないため、酵素活性、細胞膜保全性、酸化還元（Redox）能などのいくつかの指標を総合することが望まれます。それぞれのアッセイには長所と限界があり、実験に偏向がもたらされることがあるので、異なる用途には異なるアプローチがしばしば求められます。

死細胞の識別

LIVE/DEAD®固定色素LIVE/DEAD®固定死細胞染料はホルムアルデヒド固定に耐えるアミン反応性色素で、細胞内免疫表現型分析などの細胞の固定と透過性増加を必要とするアッセイを行なう前に、集団中の死細胞の「スナップショット」を画像取得することができます。細胞膜が痛んだ細胞では、色素は細胞内部と細胞表面上の両者の遊離アミンと反応し、染色され強力な蛍光を示します。生存細胞では色素の反応は細胞表面上のアミンに限定されるので、蛍光強度は低くなります。生存細胞集団と死細胞集団との間の強度の差は通常、50倍を超え、この蛍光強度の区別は病原体を不活性化させる条件を用いるホルムアルデヒド固定後も完全に保存されます。これらの単色アッセイはフローサイトメトリーの1チャンネルしか使用しないので、多重染色にも使用できます。このアッセイは蛍光顕微鏡で死細胞を検出するために用いることもできますが、生存細胞と死細胞の蛍光強度の差は感知できても2種類の集団を画像上、比較分析するのは比較的困難です。

核酸染料

SYTOX®は、細胞膜の痛んだ細胞には容易に透過する親和性の高い核酸染料ですが、生存細胞の細胞膜は通過しません。4種類のSYTOX®はそれぞれ1種類のレーザーでのみ励起するので、多重染色に使用できます。死細胞識別剤（DCD）は、フローサイトメトリー分析に供される未固定の有核細胞の細胞懸濁液で使用するように調製されています。これらの条件下において、DCDを用いて細胞膜の傷んだ非生存細胞と細胞膜が健全な生存細胞とを区別することができます。

エステラーゼ活性を検出するカルセインAMエステル

　蛍光色素のアセトキシメチル(AM)エステルは細胞内エステラーゼの優れた基質です。バイタリティ（酵素活性を介する）と膜完全性の両方の検出にこの色素AMエステルが有効です。AM基は電気的に中性もしくはほぼ中性の分子で、ほとんどの細胞に自由に色素を拡散させます。細胞内にひとたび入ると、この非蛍光エステラーゼ基質は、非特異的細胞内エステラーゼによって蛍光を発する産物に変換されます。この蛍光産物は原形質膜が損傷を受けていない細胞に保持されます。一方、酵素活性が低下した細胞および/あるいは膜が損傷を受けた細胞は、加水分解していない基質と加水分解した産物のどちらも保持しません。生存能力を有する細胞からの色素消失率は細胞のタイプに大きく依存していますが、低いインキュベーション温度と、高電荷のエステラーゼ産物は、多くの場合は保持に有利です。

　InvitrogenのLIVE/DEAD®生存率/細胞毒性キットに含まれる試薬は、生細胞群と死細胞群を2色に区別することができます。膜透過性カルセインAMは、生細胞の細胞内エステラーゼによって切断され、細胞質は緑色の蛍光を発します。膜が損傷した死細胞は、膜非透過性核酸色素、エチジウムホモダイマー -1で染色します。～ 495 nmで励起すると、カルセインAMとエチジウムホモダイマー -1から発せられる蛍光は、それぞれ～ 515 nmと～ 635 nmに区別されます。このアッセイは1時間以内で終了します。また、洗浄する必要がなく、蛍光マイクロプレートリーダーと蛍光顕微鏡検査に対応し、フローサイトメーターの使用に容易に適応できます。

精子細胞の生存率測定

セルベースアッセイ / エンドサイトーシスと貪食作用

貪食用インジケータ－pH感受性色素pHrodo™ とBioParticles®コンジュゲート

pHrodo™は酸性の貪食細胞の中だけで蛍光を発し、シグナル対ノイズ比が高く、付着しない細胞の読み取り値が改善し、正確な貪食アッセイができます。pHrodo™は細胞外で蛍光を発しないため、洗浄ステップとクエンチャー染料が必要なく、プロトコールが単純で迅速に行えます。洗浄ステップとクエンチャーがなくなることで、実験間の再現性が向上します。また、pHrodo™は柔軟な応用が可能です。pHrodo™ E.coli BioParticles®コンジュゲートはイメージングやフローに応用でき、pHrodo™活性型サクシニミジルエステル（SE）は微生物やタンパクの標識に利用できます。pHrodo™コンジュゲートは96ウェルプレートリーダーを使用することができ、蛍光顕微鏡イメージングやフローサイトメトリへの応用にも適応しています。

細胞内活性酸素生成

　InvitrogenのFc OxyBURST® Greenアッセイ試薬は、フローサイトメトリーとマイクロプレート分析により、食細胞における酸化的バースト(活性酸素発生)をモニターするために開発されました。Fc OxyBURST® Greenアッセイ試薬はジクロロジヒドロフルオレセイン(H2DCF)と共有結合させたウシ血清アルブミン(BSA)から構成されており、精製ウサギ抗BSAポリクローナル抗体と複合体を形成します。これらの免疫複合体がBSA抗体のFc領域を介してFc受容体に結合すると、非蛍光H2DCF分子が食胞内に取り込まれ、酸化されて緑色蛍光産物になります。

Fc受容体を介した食作用経路を蛍光検出するFc OxyBURST®試薬。ジクロロジヒドロフルオレセイン(H2DCF)がBSAと共有結合しており、ウサギ抗BSAポリクローナル抗体と複合体を形成する。Fc受容体に結合すると非蛍光免疫複合体は取り込まれて酸化され、蛍光DCFになる。

健常ドナー（コントロール）とヒト慢性肉芽腫症（CGD）患者からの好中球の活性酸素発生の比較。Fc OxyBURST®グリーンアッセイ試薬を使用して検出した。フローサイトメトリーのデータは、Boston UniversityのElizabeth
